Digital dermatitis is a highly prevalent painful lesion affecting the feet in dairy cattle. Even though the pathogenesis has been subject of investigation since 1974, there is still a lack of knowledge about the spread of the disease among cows within a herd as well as between herds. The purpose of this study was to monitor transmission of digital dermatitis under experimental conditions between naive heifers and affected animals, to monitor the changes in clinical appearance, microbial colonisation of the skin as lesions progressed and to apply a q-PCR for the detection of Treponema spp. in faecal samples. Eight heifers with clinical normal digital skin were housed with 5 heifers with severe digital dermatitis lesion for 8 weeks on a solid concrete floor with an accumulating layer of slurry. Digital skin was examined daily and lesions were clinically scored. Skin biopsies were taken from the healthy heifers at introduction and weekly from all lesions for histopathological evaluation and fluorescence in situ hybridization. None of the healthy heifers developed digital dermatitis and in 4 out of 5 infected heifers the lesions healed during the study. All samples from healthy skin were negative for Treponema spp. and one sample were positive for Dichelobacter nodosus. Colonization of healthy skin could not be identified in this study. There was no significant relation between clinical scoring of the lesions and histopathological score and the presence of Treponema spp. There were however a significant relation between the prevalence of Treponema spp. in the skin and severity of changes in epidermis and dermis. By qPCR all the healthy heifers were found to excrete Treponema spp. in their faeces.

The objective of these studies was to investigate if porcine postweaning multisystemic wasting syndrome (PMWS) could be induced in healthy pigs following contact with air from pigs with clinical signs of PMWS. The pigs were housed in different units. Either 31 (study I) or 25 (study II) pigs with clinical symptoms of PMWS from a PMWS-affected herd and 25 healthy pigs from a PMWS-free, but PCV2-positive, herd were housed in unit A. Fifty pigs from a PMWS-free herd were housed in unit B, which were connected by pipes to unit A. In unit C, 30 pigs from a PMWS-free herd were housed as controls. In study II, the pigs in units A and B from the PMWS-free herd developed clinical signs of PMWS 2-3 weeks after arrival. PMWS was confirmed at necropsy and the diseased pigs had increased PCV2 load and increased antibody titers against PCV2 in serum that coincided with the development of clinical signs typical of PMWS. Sequence analysis revealed that the PCV2 isolate belonged to genotype 2b. In conclusion, the present study showed that PMWS can be induced in pigs from a PMWS-free herd by airborne contact with pigs from a PMWS-affected herd.

The effects of four types of plastic surfaces and four pre-incubation media, containing high/low glucose and +/- amino acids, on adhesion of Saccharomyces cerevisiae BY4742 wild type and Deltaflo11 mutant (strain background S288c) were investigated. No difference in adhesive ability between the two yeast strains was observed in any of our experiments, thus confirming that FLO11 is not operational in the S. cerevisiae S288c strain background. The adhesive abilities of both yeast strains depended on the plastic type and pre-incubation conditions. The poorest adhesion was observed on hydrophilic polystyrene, whereas hydrophobic polystyrene resulted in moderate adhesion. The best adhesion of both yeast strains was observed on polystyrene surfaces with combined hydrophilic/hydrophobic domains. When amino acids were present in the pre-incubation media, lack of glucose increased the cell surface hydrophobicity and enhanced the adhesion to all four types of polystyrene. Lack of amino acids in the pre-incubation media increased the cell surface hydrophobicity and enhanced the adhesion especially to polystyrene surfaces with combined hydrophilic/hydrophobic domains. Our results suggest that glucose and amino acid starvation induces other genes than FLO11 in S. cerevisiae S288c coding for hydrophobic cell surface constituents with adhesive properties to especially moderately hydrophobic plastic surfaces.
