A biodegradable hybrid polymer patch was invented at the University of Cincinnati to cover gaps on the skin over the spinal column of a growing fetus, characterized by the medical condition spina bifida. The inserted patch faces amniotic fluid (AF) on one side and cerebrospinal fluid on the other side. The goal is to provide a profile of the roughness of a patch over time at 0, 4, 8, 12, and 16 weeks with a 95% confidence band. The patch is soaked in a test tube filled with either amniotic fluid (AF) or phosphate-buffered saline (PBS) in the lab. If roughness is measured at any time point for a patch, the patch is destroyed. Thus, it is impossible to measure roughness at all weeks of interest for any patch. It is important to assess the roughness of a patch because the rougher the patch is, the faster the skin grows under the patch. We use a model-based approach with Monte Carlo simulations to estimate the profile over time with a 95% confidence band. The roughness profiles are similar with both liquids. The profile can be used as a template for future experiments on the composition of patches.

Leaf area index (LAI) is a canopy structure parameter used to characterize vegetation growth. It is increasingly important to obtain continuous and accurate ground LAI measurement data for the validation of remote-sensing products. In this study, direct destructive sampling method and indirect optical measurement including LAI-2200 plant canopy analyser (PCA) and Tracing Radiation and Architecture of Canopies (TRAC) were used for long-term continuous observation of paddy fields in southern China, to explore the variation of LAI during the growth period of paddy, and to quantitatively analyse the errors of indirect optical measurement. The results show that (1) LAI and Plant area index (PAI) increased first and then decreased in the growing period. The proportion of green leaf area gradually decreased at reproductive growth stage (less than 50%). There was a linear relationship between the proportion of green leaf area and PAI at this stage, with a good correlation (R2 = 0.73). This linear relationship can be used to convert PAI into LAI more conveniently. (2) Effective PAI (PAIe) calculated by the two methods (2000 method and Lang method) from PCA showed good consistency between each other in the growing season (the maximum value of R2 is 0.99). With the increase of zenith angle, the consistency between 2000 method and Lang method decreases (R2 = 0.83). Multiple scattering has an effect on PCA measurement, and this effect is more obvious at high zenith angle. Lang method is more sensitive to the change of zenith angle. (3) Compared to the destructive values, both 2000 method and Lang method are able to compute accurate LAI using four ring sensors (4 R), while TRAC underestimates the LAI and PAI from destructive sampling method. A commingled waste rock and tailings test pile and a waste rock test pile were evaluated to determine saturated hydraulic conductivity and destructively sampled to measure dry density. The commingled test pile contained a mixture of filtered tailings and waste rock blended to isolate waste rock particles as inclusions within the tailings matrix. Test piles were constructed in the shape of truncated 5 m high pyramids with 25 m base sides and flat 5 m x 5 m top surfaces, and instrumented to monitor water content (and additional geochemical indicator parameters) within the test pile and seepage from the base of the pile. Piles were decommissioned after 26 months of operation. Saturated hydraulic conductivities were measured using sealed double-ring infiltrometers (2.4 m square outer ring and 1 m square inner ring). Tensiometers and embedded water content sensors were used to measure progression of the wetting front, and the final location of the wetting front in the commingled test pile was directly measured during decommissioning. Field-measured saturated hydraulic conductivities were compared to laboratory-measured results intended to simulate the test piles. Despite having a lower average density, the commingled waste rock and tailings had a hydraulic conductivity approximately 2.5 times lower than the waste rock. Ils etaient equipes d'instruments permettant de surveiller la teneur en eau (et d'autres parametres indicateurs geochimiques) dans le pieu d'essai et le suintement a partir de la base du pieu. Les pieux ont ete mis hors service apres 26 mois d'exploitation. On a mesure les conductivites hydrauliques saturees par le biais d'infiltrometres scelles a double anneau (anneau exterieur carre de 2,4 m et anneau interieur carre de 1 m). La progression du front d'humectation a ete mesuree a l'aide de tensiometres et de capteurs de teneur en eau integres, et l'emplacement final du front d'humectation dans le pieu d'essai melange a ete mesure directement pendant le demantelement. Les conductivites hydrauliques saturees mesurees sur le terrain ont ete comparees aux resultats mesures en laboratoire pour simuler les pieux d'essai. Evaluating the genetic status of threatened species is an essential task in conservation genetics. However, the genetic status of threatened species has been mostly evaluated through techniques that fail to estimate genetic diversity at the whole genomic level. Next generation sequencing can meet this demand, but high quality samples such as blood or muscle tissues are required. However, it is difficult to collect such samples from threatened species because sampling work may impact their health. Therefore, it is essential to design a workflow to evaluate the whole genomic status of threatened species using non-destructive sampling. Even though non-destructive sampling has been used in traditional barcoding technique, the barcoding technique cannot evaluate the whole genomic status. Brown Eared Pheasant (Crossoptilon mantchuricum) is an endangered species, with captive populations maintained in Taiyuan Zoo, China, and Europe. However, the genetic diversity, inbreeding pattern, and mutation load of these two populations are unclear. To uncover the genetic status of these two captive populations, we applied 2b-RAD technology to evaluate the genomic status of these populations using feathers as samples. The feathers could be collected by non-destructive sampling. The results indicate that the Taiyuan Zoo population has a lower genetic diversity and higher inbreeding coefficient than the European population. The Taiyuan Zoo population has lethal mutations when homozygous. The current project uses a non-destructive sampling technique to evaluate the whole genomic status of the two captive populations, providing a paradigm for conservation genetics, which will facilitate the development of conservation biology.

Parchment, the skins of animals prepared for use as writing surfaces, offers a valuable source of genetic information. Many have clearly defined provenance, allowing for the genetic findings to be evaluated in temporal and spatial context. While these documents can yield evidence of the animal sources, the DNA contained within these aged skins is often damaged and fragmented. Previously, genetic studies targeting parchment have used destructive sampling techniques and so the development and validation of non-destructive sampling methods would expand opportunities and facilitate testing of more precious documents, especially those with historical significance. Here we present genetic data obtained by non-destructive sampling of eight parchments spanning the 15th century to the modern day. We define a workflow for enriching the mitochondrial genome (mtGenome), generating next-generation sequencing reads to permit species identification, and providing interpretation guidance. Using sample replication, comparisons to destructively sampled controls, and by establishing authentication criteria, we were able to confidently assign full/near full mtGenome sequences to 56.3% of non-destructively sampled parchments, each with greater than 90% of the mtGenome reference covered. Six of eight parchments passed all four established thresholds with at least one non-destructive sample, highlighting promise for future studies. [ABSTRACT FROM AUTHOR]

With the advent of ancient DNA, as well as other methods such as isotope analysis, destructive sampling of archaeofaunal remains has increased much faster than the effort to collect and curate them. While there has been considerable discussion regarding the ethics of destructive sampling and analysis of human remains, this dialogue has not extended to archaeofaunal material. Here we address this gap and discuss the ethical challenges surrounding destructive sampling of materials from archaeofaunal collections. We suggest ways of mitigating the negative aspects of destructive sampling and present step-by-step guidelines aimed at relevant stakeholders, including scientists, holding institutions and scientific journals. Our suggestions are in most cases easily implemented without significant increases in project costs, but with clear long-term benefits in the preservation and use of zooarchaeological material.

This study aimed to determine the wood density using destructive and non-destructive sampling methods in a eucalypt hybrid clonal and determine the best point for the non-destructive sampling of the tree. A 5.6-year-old Eucalyptus urophylla x Eucalyptus grandishybrid clone, from an energetic forest, was used, planted in the municipal district of Martinho Campos, MG, Brazil. The trees were sampled by three methods of destructive sampling. The traditional method (0%, 25%, 50%,75% and 100% of the commercial height Hc); alternative method (2%, 10%, 30% and 70% Hc) and via a meter by meter method starting from a height of 1.30 m from the soil (DBH). Three non-destructive sampling methods were also appraised: Resistograph®, Pilodyn® and 22 mm diameter cores at a DBH. The use of the Resistograph® at the 1.10 m or 1.50 m points and the Pilodyn® at 1.50m of height from the soil results the best correlations with the basic density determined by the three destructive sampling methods considered.

The study of volatile compounds obtained by gas chromatography (GC) coupled to mass spectrometry (MS) or ion mobility spectrometry (IMS) may be very useful to protect food quality, especially when using a non-destructive sampling method. In this work, the authentication of the highly appreciated dry-cured Iberian ham by those techniques was studied and compared. The results obtained show the suitability of a non-destructive sampling method coupled to headspace sampling (HS)-GC-IMS or HS-GC-MS to determine volatile markers in the feeding Iberian pig regime. Although both methods were suitable to differentiate the ham categories, HS-GC-IMS was more sensitive detecting a higher number of compounds than HS-GC-MS, which provided accurate qualitative results. The results of principal component analysis showed that ethanol, 2-propanol and 3-methylbutanol, identified by HS-GC-IMS, and 3-methylbutanal and heptane, identified by HS-GC-MS, could be considered potential markers to identify ham from different feeding regimes., (Copyright © 2021 Elsevier Ltd. Abstract The estimation and monitoring of the huge amount of carbon contained in tropical forests, and specifically in the above-ground biomass (AGB) of trees, is needed for the successful implementation of climate change mitigation strategies. Its accuracy depends on the availability of reliable allometric equations to convert forest inventory data into AGB estimates. In this study, we tested whether central African forests are really different from other tropical forests with respect to biomass allometry, and further examined the regional variation in tropical tree allometry across the Congo basin forests. Following the same standardized protocol, trees were destructively sampled for AGB in six sites representative of terra firme forests. We fitted regional and local allometric models, including tree diameter, wood specific gravity, tree height, and crown radius in the AGB predictors. We also evaluated the AGB predictions at the tree level across the six sites of our new models and of existing allometric models, including the pantropical equations developed by Chave et al. (2014, 2005) and the local equations developed by Ngomanda et al. (2014) in Gabon. With a total of 845 tropical trees belonging to 55 African species and covering a large range of diameters (up to 200 cm), the original data presented here can be considered as the largest ever destructive sampling for a tropical region. Regional allometric models were established and including tree height and crown radius had a small but significant effect on AGB predictions. In contrast to our expectations, tree height and crown radius did not explain much between-site variation. Examining the performance of general models (pantropical or regional) versus local models (site-specific), we found little advantage of using local equations. Earlier pantropical equations developed for moist forests were found to provide reasonable predictions of tree AGB in most sites, though the wettest sites, i.e. , evergreen forests in Equatorial Guinea and, to a lesser extent in Gabon, tended to show a wet forest allometry. For the Congo basin forests, except in Equatorial Guinea where local models might be preferred, we recommend using our regional models, and otherwise the most recent pantropical models, that were validated here. These results constitute a critical step for the estimation and monitoring of biomass/carbon stocks contained in the second largest contiguous block of tropical forests worldwide, and the successful implementation of climate change mitigation strategies, such as REDD+.
