Abstract

The unification of quantum mechanics and general relativity has long been elusive. Only recently have empirical predictions of various possible theories of quantum gravity been put to test. The dawn of multi-messenger high-energy astrophysics has been tremendously beneficial, as it allows us to study particles with much higher energies and travelling much longer distances than possible in terrestrial experiments, but more progress is needed on several fronts. A thorough appraisal of current strategies and experimental frameworks, regarding quantum gravity phenomenology, is provided here. Our aim is twofold: a description of tentative multimessenger explorations, plus a focus on future detection experiments. As the outlook of the network of researchers that formed through the COST Action CA18108 "Quantum gravity phenomenology in the multi-messenger approach (QG-MM)", in this work we give an overview of the desiderata that future theoretical frameworks, observational facilities, and data-sharing policies should satisfy in order to advance the cause of quantum gravity phenomenology., Comment: Submitted to CQG for the Focus Issue on "Quantum Gravity Phenomenology in the Multi-Messenger Era: Challenges and Perspectives". Abstract

The origin of fluorine is a widely debated issue. Nevertheless, the ^{15}N({\alpha},{\gamma})^{19}F reaction is a common feature among the various production channels so far proposed. Its reaction rate at relevant temperatures is determined by a number of narrow resonances together with the DC component and the tails of the two broad resonances at E_{c.m.} = 1323 and 1487 keV. Measurement through the direct detection of the 19F recoil ions with the European Recoil separator for Nuclear Astrophysics (ERNA) were performed. The reaction was initiated by a 15N beam impinging onto a 4He windowless gas target. The observed yield of the resonances at Ec.m. = 1323 and 1487 keV is used to determine their widths in the {\alpha} and {\gamma} channels. We show that a direct measurement of the cross section of the ^{15}N({\alpha},{\gamma})^{19}F reaction can be successfully obtained with the Recoil Separator ERNA, and the widths {\Gamma}_{\gamma} and {\Gamma}_{\alpha} of the two broad resonances have been determined. While a fair agreement is found with earlier determination of the widths of the 1487 keV resonance, a significant difference is found for the 1323 keV resonance {\Gamma}_{\alpha} . The revision of the widths of the two more relevant broad resonances in the 15N({\alpha},{\gamma})19F reaction presented in this work is the first step toward a more firm determination of the reaction rate. At present, the residual uncertainty at the temperatures of the ^{19}F stellar nucleosynthesis is dominated by the uncertainties affecting the Direct Capture component and the 364 keV narrow resonance, both so far investigated only through indirect experiments., Comment: 8 pages, 11 figures. Abstract

Motivation: Target enrichment strategies generate genomic data from multiple pathogens in a single process, greatly improving sensitivity over metagenomic sequencing and enabling cost-effective, high-throughput surveillance and clinical applications. However, uptake by research and clinical laboratories is constrained by an absence of computational tools that are specifically designed for the analysis of multi-pathogen enrichment sequence data. Here we present an analysis pipeline, Castanet, for use with multi-pathogen enrichment sequencing data. Castanet is designed to work with short-read data produced by existing targeted enrichment strategies, but can be readily deployed on any BAM file generated by another methodology. Also included are an optional graphical interface and installer script., Results: In addition to genome reconstruction, Castanet reports method-specific metrics that enable quantification of capture efficiency, estimation of pathogen load, differentiation of low-level positives from contamination, and assessment of sequencing quality. Castanet can be used as a traditional end-to-end pipeline for consensus generation, but its strength lies in the ability to process a flexible, pre-defined set of pathogens of interest directly from multi-pathogen enrichment experiments. In our tests, Castanet consensus sequences were accurate reconstructions of reference sequences, including in instances where multiple strains of the same pathogen were present. Abstract

The effect of torsion in theories of quantum gravity is known to be well described by an axion-like field which couples to matter as well as to gravitation and radiation gauge fields. In this note we consider a particular kind of torsion, arising from the Kalb-Ramond antisymmetric tensor field that appears in the gravitational multiplet of string theory. We investigate the implications for leptogenesis. Abstract

The assessment of the radiological impact of a decommissioning Nuclear Power Plant is presented here through the results of an environmental monitoring survey carried out in the area surrounding the Garigliano Power Plant. The levels of radioactivity in soil, water, air and other environmental matrices are shown, in which {\alpha}, {\beta} and {\gamma} activity and {\gamma} equivalent dose rate are measured. Radioactivity levels of the samples from the Garigliano area are analyzed and then compared to those from a control zone situated more than 100 km away. Moreover, a comparison is made with a previous survey held in 2001. The analyses and comparisons show no significant alteration in the radiological characteristics of the area surroundings the plant, with an overall radioactivity depending mainly from the global fallout and natural sources., Comment: 13 pages, 6 figures, 2 tables

Abstract

The paper offers a contribution to the study of Lilibeo’s figured vases, which make extensive use of added color. The examination of some contexts allows us to clarify the old idea of a derivation of the Lilibetane vases from the workshops of Lipari, due to a displacement of artisans who would operate first in Lipari and then in Lilibeo (like the Swans Painter). It is not completely excludes the movement of craftsmen due to destructions or political changes. However it reconstructs complex interactions in which a main role had to play the influence and the action of medium exercised by the same imported artifacts (Lipari vases, mainly, but not only).

Abstract

Over the last years, automotive industries drove a great amount of research in the field of advanced combustion techniques minimizing carbon dioxide emissions. The so-called Low-Temperature Combustions (LTC), characterized by the self-ignition of highly premixed air-fuel mixtures, represent a promising solution to achieving high efficiency and ultralow emissions of nitrogen oxides (NOx) and particulate matter. Among these, gasoline Partially Premixed Combustion (PPC), obtained through the high-pressure direct injections of gasoline, showed a good potential for the simultaneous reduction of pollutants and emissions in compression ignited engines. However, when multiple injections per cycle are performed (with hydraulic-assisted needle opening), combustion stability might be compromised by the wave effects in the hydraulic system, which produce incoherence between the requested and injected fuel. This work presents a model-based pressure waves reconstruction strategy, based on a control-oriented model of the high-pressure common rail injection system fueled with gasoline. To determine the hydraulic system's behavior during the injection process, a specifically designed flushing bench with a high-frequency acquisition system has been developed. Experimental activities have been carried out to highlight fuel pressure fluctuations with single and double injection patterns. Through the analysis of the acquired data, the key parameters (characteristic of the system) have been identified and the accuracy of pressure waves reconstruction has been evaluated, always returning errors lower than 2% between measured and estimated instantaneous pressures. Different fuel types, injectors, and rail positions have been tested to highlight the robustness of the approach. Based on the instantaneous pressure trace estimated with the control-oriented model, a fuel quantity Fluctuation Correction Strategy (FQC), implementable on a standard engine Electronic Control Unit (ECU), has been developed. The obtained results confirm the potential to reduce fuel quantity oscillations in multiple-injections systems.

Abstract

Diffuse malignant peritoneal mesothelioma (DMPM) is a rare and rapidly lethal tumor, poorly responsive to conventional treatments. In this regards, the identification of molecular alterations underlying DMPM onset and progression might be exploited to develop novel therapeutic strategies. Here, we focused on miR-550a-3p, which we found downregulated in 45 DMPM clinical samples compared to normal tissues and whose expression levels were associated with patient outcome. Through a gain-of-function approach using miRNA mimics in 3 DMPM cell lines, we demonstrated the tumor-suppressive role of miR-550a-3p. Specifically, miRNA ectopic expression impaired cell proliferation and invasiveness, enhanced the apoptotic response, and reduced the growth of DMPM xenografts in mice. Antiproliferative and proapoptotic effects were also observed in prostate and ovarian cancer cell lines following miR-550a-3p ectopic expression. miR-550a-3p effects were mediated, at least in part, by the direct inhibition of HSP90AA1 and the consequent downregulation of its target proteins, the levels of which were rescued upon disruption of miRNA-HSP90AA1 mRNA pairing, partially abrogating miR-550a-3p-induced cellular effects. Our results show that miR-550a-3p reconstitution affects several tumor traits, thus suggesting this approach as a potential novel therapeutic strategy for DMPM., (© 2022. The Author(s).)

Abstract

Respiratory syncytial virus (RSV) is the leading cause of hospitalisation for respiratory infection in young children. RSV disease severity is known to be age-dependent and highest in young infants, but other correlates of severity, particularly the presence of additional respiratory pathogens, are less well understood. In this study, nasopharyngeal swabs were collected from two cohorts of RSV-positive infants <12 months in Spain, the UK, and the Netherlands during 2017-20. We show, using targeted metagenomic sequencing of >100 pathogens, including all common respiratory viruses and bacteria, from samples collected from 433 infants, that burden of additional viruses is common (111/433, 26%) but only modestly correlates with RSV disease severity. In contrast, there is strong evidence in both cohorts and across age groups that presence of Haemophilus bacteria (194/433, 45%) is associated with higher severity, including much higher rates of hospitalisation (odds ratio 4.25, 95% CI 2.03-9.31). There is no evidence for association between higher severity and other detected bacteria, and no difference in severity between RSV genotypes. Our findings reveal the genomic diversity of additional pathogens during RSV infection in infants, and provide an evidence base for future causal investigations of the impact of co-infection on RSV disease severity., (© 2024. Abstract

Genomic surveillance of Plasmodium falciparum malaria can provide policy-relevant information about antimalarial drug resistance, diagnostic test failure, and the evolution of vaccine targets. Yet the large and low complexity genome of P. falciparum complicates the development of genomic methods, while resource constraints in malaria endemic regions can limit their deployment. Here, we demonstrate an approach for targeted nanopore sequencing of P. falciparum from dried blood spots (DBS) that enables cost-effective genomic surveillance of malaria in low-resource settings. We release software that facilitates flexible design of amplicon sequencing panels and use this software to design two target panels for P. falciparum. The panels generate 3-4 kbp reads for eight and sixteen targets respectively, covering key drug-resistance associated genes, diagnostic test antigens, polymorphic markers and the vaccine target csp. We validate our approach on mock and field samples, demonstrating robust sequencing coverage, accurate variant calls within coding sequences, the ability to explore P. falciparum within-sample diversity and to detect deletions underlying rapid diagnostic test failure., (© 2024. Abstract

Background: In the last decade, universally available antiretroviral therapy (ART) has led to greatly improved health and survival of people living with HIV in sub-Saharan Africa, but new infections continue to appear. The design of effective prevention strategies requires the demographic characterisation of individuals acting as sources of infection, which is the aim of this study., Methods: Between 2014 and 2018, the HPTN 071 PopART study was conducted to quantify the public health benefits of ART. Viral samples from 7124 study participants in Zambia were deep-sequenced as part of HPTN 071-02 PopART Phylogenetics, an ancillary study. We used these sequences to identify likely transmission pairs. After demographic weighting of the recipients in these pairs to match the overall HIV-positive population, we analysed the demographic characteristics of the sources to better understand transmission in the general population., Findings: We identified a total of 300 likely transmission pairs. 178 (59·4%) were male to female, with 130 (95% CI 110-150; 43·3%) from males aged 25-40 years. Overall, men transmitted 2·09-fold (2·06-2·29) more infections per capita than women, a ratio peaking at 5·87 (2·78-15·8) in the 35-39 years source age group. 40 (26-57; 13·2%) transmissions linked individuals from different communities in the trial. Of 288 sources with recorded information on drug resistance mutations, 52 (38-69; 18·1%) carried viruses resistant to first-line ART., Interpretation: HIV-1 transmission in the HPTN 071 study communities comes from a wide range of age and sex groups, and there is no outsized contribution to new infections from importation or drug resistance mutations. Abstract

Durability tests of thermal protection system (TPS) materials for space vehicles is of paramount importance in developing TPS materials. The limits of the presently used techniques and the need for higher accuracy and precision demand new reliable and more sensitive measurement methods. In this paper we present the validation of a new on-line and non intrusive technique to estimate the TPS material recession rate which exploits the surface layer implantation of radioactive Be-7 ions and the measurement of its characteristic gamma-rays during the recession test.

Abstract

The hepatitis C virus (HCV) epidemic in Western countries is primarily perpetuated by the sub-populations of men who have sex with men (MSM) and people who inject drugs (PWID). Understanding the dynamics of transmission in these communities is crucial for removing the remaining hurdles towards HCV elimination. We sequenced 269 annotated HCV plasma samples using probe enrichment and next-generation sequencing, obtaining 224 open reading frames of HCV (OR497849-OR498072). Maximum likelihood phylogenies were generated on the four most prevalent subtypes in this study (HCV1a, 1b, 3a, 4d) with a subsequent transmission cluster analysis. The highest rate of clustering was observed for HCV4d samples (13/17 (76.47%)). The second highest rate of clustering was observed in HCV1a samples (42/78 (53.85%)) with significant association with HIV-positive MSM. HCV1b and HCV3a had very low rates of clustering (2/83 (2.41%) and (0/29)). The spread of the prevalent subtype HCV1b appears to have been largely curtailed, and we demonstrate the onwards transmission of HCV1a and HCV4d in the HIV-positive MSM population across municipal borders. More systematic data collection and sequencing is needed to allow a better understanding of the HCV transmission among the community of PWID and overcome the remaining barriers for HCV elimination in Belgium.

Abstract

The spike protein of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is critical for virus infection through the engagement of the human ACE2 protein1 and is a major antibody target. Here we show that chronic infection with SARS-CoV-2 leads to viral evolution and reduced sensitivity to neutralizing antibodies in an immunosuppressed individual treated with convalescent plasma, by generating whole-genome ultra-deep sequences for 23 time points that span 101 days and using in vitro techniques to characterize the mutations revealed by sequencing. There was little change in the overall structure of the viral population after two courses of remdesivir during the first 57 days. However, after convalescent plasma therapy, we observed large, dynamic shifts in the viral population, with the emergence of a dominant viral strain that contained a substitution (D796H) in the S2 subunit and a deletion (ΔH69/ΔV70) in the S1 N-terminal domain of the spike protein. As passively transferred serum antibodies diminished, viruses with the escape genotype were reduced in frequency, before returning during a final, unsuccessful course of convalescent plasma treatment. In vitro, the spike double mutant bearing both ΔH69/ΔV70 and D796H conferred modestly decreased sensitivity to convalescent plasma, while maintaining infectivity levels that were similar to the wild-type virus.The spike substitution mutant D796H appeared to be the main contributor to the decreased susceptibility to neutralizing antibodies, but this mutation resulted in an infectivity defect. The spike deletion mutant ΔH69/ΔV70 had a twofold higher level of infectivity than wild-type SARS-CoV-2, possibly compensating for the reduced infectivity of the D796H mutation. These data reveal strong selection on SARS-CoV-2 during convalescent plasma therapy, which is associated with the emergence of viral variants that show evidence of reduced susceptibility to neutralizing antibodies in immunosuppressed individuals.

Abstract

The paper aims to reassess the entire architectural and archaeological evidence of the southern part of the sanctuary of Zeus Olympios. This includes the oikos near the SE corner of the big temple (with two building phases dated respectively to the mid-6th century and to the first half of the 5th century), and the area with a large pool and two halls located to the south. This is equipped with a complex drainage system connected with two big cisterns. All these structures can now be referred to as a coherent building program carried out from 480 to 450 B.C. The monumentality of the buildings and infrastructures reflects a grandeur of the rituals that has not been previously addressed. Until now, most scholars have focused only on the colossal Temple of Zeus, and the sanctuary was not perceived as a whole. On the contrary, it was a structured and organic ensemble that could fulfil the multiple needs of religious and festive occasions. At the same time, it engaged in dialogue with the other components of the ‘sacred acropolis’, from the ‘Herakles Temple’ to the sanctuaries to the west. As for the rites and cults practiced in this area, the findings related mainly to the last phase of usage of the sacellum suggest a female divinity, with a sphere of activities linked to female maturation. The large swimming pool and the adjoining open-air area were an integral part of these rituals. To the same group of structures belongs the next monumental hall; a stone structure facing the main entrance (a big kline or trapeza) suggests the performance of theoxenic rituals. It is an intriguing coincidence that Pindar describes how the Tyndaridae were honoured with a special emphasis by the same Teron, to whom the project of the Zeus Olympios sanctuary can be attributed.

Abstract

A seguito delle ricerche idell’Università degli studi di Palermo, si propone una nuova lettura d’insieme dell’area sacra situata sul margine meridionale della città di Akragas, finora percepita sostanzialmente attraverso i templi monumentali più noti (Olympieion e cosiddetto Tempio di Ercole) ma in realtà frutto di una complessa sequela di interventi di infrastrutturazione e riconfigurazione monumentale, legati all’evoluzione socio-politica e alla crescita economico-materiale della città dal suo sorgere alla conquista romana. Following research by the University of Palermo, a new overview of the sacred area located on the southern edge of the city of Akragas is proposed, so far perceived substantially through the best-known monumental temples (Olympieion and so-called Temple of Hercules) but in reality the result of a complex sequence of interventions of infrastructure and monumental reconfiguration, linked to the socio-political evolution and the economic-material growth of the city from its rise to the Roman conquest.

Il contributo prende in considerazione i crateri-cinerari figurati restituiti dalle necropoli greche di Sicilia, ricollegati, in via di ipotesi, a credenze escatologiche connesse alla sfera del culto dionisiaco, cercando di rintracciare l’origine e di inquadrare l’affermarsi di tale rito in Sicilia, e di delineare gli attori di questo rituale, con il quale l’identità del/della defunto/a era richiamata dalla forma del cinerario e dalle immagini che lo decoravano. L’analisi prende avvio dai cinerari con raffinate e complesse scene mitiche dell’Agrigento dell’età della tirannide emmenide per arrivare ai crateri della Lipara del IV secolo, che esprimono attraverso narrazioni più esplicite l’auspicio (o la certezza) di un accesso a una dimensione salvifica e di felicità eterna, di un accoglimento post mortem nel thiasos divino. Si ricostruisce una pluralità di segni e mezzi espressivi variamente declinati che arricchisce di sfumature e articola il contesto socio-culturale di riferimento e il relativo panorama rituale nello spazio e nel tempo, pur in un comune sottofondo di credenze escatologiche, maturate e cresciute probabilmente nella Sicilia delle tirannidi, e sviluppatesi poi in maniera originale ma coerente nei singoli contesti, nei diversi tessuti sociali, nella trama delle differenti ritualità funerarie nel secolo successivo e oltre, quando il mito perderà a poco a poco la sua funzione legittimante e identitaria, soppiantato da narrazioni più dichiaratamente misteriche e iniziatiche. The contribution takes into consideration the figured cinerary craters returned from the Greek necropolises of Sicily, linked, by way of hypothesis, to eschatological beliefs connected to the sphere of the Dionysian cult, trying to trace the origin and to frame the affirmation of this rite in Sicily, and to outline the actors of this ritual, with which the identity of the deceased was recalled by the shape of the cinerary and by the images that decorated it. The analysis starts from the cineraries with refined and complex mythical scenes of Agrigento from the age of the emmenid tyranny to arrive at the craters of Lipara of the fourth century, which express through more explicit narratives the hope (or the certainty) of access to a saving dimension and eternal happiness, of a post mortem acceptance in the divine thiasos. A plurality of variously declined signs and expressive means is reconstructed that enriches with nuances and articulates the socio-cultural context of reference and the relative ritual panorama in space and time, albeit in a common background of eschatological beliefs, probably matured and grown in Sicily tyrannies, and then developed in an original but coherent way in the individual contexts, in the different social tissues, in the plot of the different funeral rituals in the following century and beyond, when the myth gradually loses its legitimizing and identity function, supplanted by narratives more openly mysterious and initiatory.
