Introduction: Emicizumab mimicking the cofactor function of activated factor VIII (FVIII) restores haemostasis. Methods: This nationwide observational study aimed to retrospectively investigate efficacy, safety, and cost in 1 year before and up to 3 years after emicizumab prophylaxis for haemophilia A (HA) patients with FVIII inhibitors. Results and discussion: A total of 39 severe HA patients with a median age of 23.0 years were enrolled. The median historical peak FVIII inhibitor titre was 174.2 BU/mL with an interquartile range of 56.5–578.8 BU/mL. The median annualized bleeding rate reduced from 24 to 0 events in the first year after emicizumab prophylaxis (p <.01) and sustained in the second and third years. The median annualized joint bleeding rate reduced to 0 and maintained up to 3 years (p <.01). Twenty‐seven patients (69.2%) had target joints before emicizumab prophylaxis and only seven patients (17.9%) of them had target joints after prophylaxis. Medical costs, including cost of haemostatic therapy, frequency of outpatient department visits, emergency room visits and hospital admission, were significantly reduced after emicizumab prophylaxis (p <.01). FVIII inhibitor titre decreased after emicizumab prophylaxis. Overall, three (7.7%) patients experienced 202 grade 1 drug‐related adverse events after emicizumab prophylaxis. No serious adverse events were reported during emicizumab prophylaxis period. The adherence to emicizumab prophylaxis was 100% up to 3 years. Conclusions: HA patients with FVIII inhibitors treated with emicizumab prophylaxis resulted in a significant reduction in treated bleeds and associated costs. No new safety events were observed. Abstract Background B7 Costimulatory signal is essential to trigger T-cell activation upon the recognition of tumor antigens. This study examined the expression of B7-1 (CD80) and B7-2 (CD86) costimulatory molecules along with HLA-DR and the presence of infiltrating lymphocytes and dendritic cells to assess their significance in patients with nasopharyngeal carcinoma (NPC). Methods Expression of CD80, CD86, HLA-DR, S-100 protein and the presence of infiltrating lymphocytes and follicular dendritic reticulum cells were immunohistochemically examined on the paraffin-embedded tissue blocks from newly diagnosed NPC patients (n = 50). The results were correlated with clinical outcome of patients. Results CD80 and CD86 were each expressed in 10 of 50 cases in which they co-expressed in 9 cases. Univariate analysis revealed that patients with CD80/CD86 expression had significantly better overall survival than those without it (P = 0.017), but after adjustment for stage, nodal status, and treatment, the expression of CD80/CD86 did not significantly correlate with overall survival. Expression of HLA-DR and the presence of infiltrating lymphocytes and dendritic cells did not appear to have impact on the survival of patients. Conclusion Expression of CD80 and CD86 costimulatory molecules appears to be a marker of better survival in patient with NPC.

In the present study, after solid solution treatment, four different artificial aging treatments (100, 120, 140 and 160 °C) were performed on Al-5.98Zn-2.86Mg-1.61Cu (wt.%) alloy, denoted as 7075-LCu, and Al-5.91Zn-2.83Mg-1.98Cu (wt.%) alloy, denoted as 7075-HCu. Peak aging conditions were determined for each aging temperature at various hold time intervals of up to 24 h. It was found that both alloys possessed the optimal strengths after artificial aging at 120 °C for 24 h. Under this condition, the ultimate tensile strengths (UTSs) were 618 MPa (7075-LCu) and 623 MPa (7075-HCu), respectively. Moreover, a method was used to calculate the average sizes and number density of the major strengthening precipitates, η′, under peak aging conditions in these two alloys from transmission electron microscopy (TEM) images and electron energy loss spectroscopy (EELS). The above results indicated that for the 7075-LCu and 7075-HCu samples with the optimal UTS strengths, the former possessed an average thickness of 2.15 nm, and a number density of 3.27 × 1017 cm−3; the latter, 2.04 nm and 3.52 × 1017 cm−3. Two steels with a base composition of Fe-0.2C-0.8Mn-1.2Cr (wt%) but with different niobium (Nb) contents (0.02 and 0.03 wt%) were employed to study the effect of precipitate evolution on the softening resistance in the austenite region under elevated temperature deformation. The thermomechanical procedure was executed by a deformation-dilatometer and involved double deformation processes with 25% strain at a 0.25 s−1 strain rate at 900, 925, 950, and 1000 °C. The softening ratios, reflecting the competition between recrystallization and precipitation, were evaluated. The results indicated that both steels showed better softening resistance at 900 °C than at other temperatures. However, the softening ratio of 0.03 wt% Nb-containing steel (Steel 3N) rose after 100 s at 900 °C, while 0.02 wt% Nb-containing steel (Steel 2N) maintained a low softening ratio within 300 s at 900 °C, indicating that Steel 3N was relatively non-durable. A microstructural characterization showed that in the Steel 3N sample deformed at 900 °C, recrystallization occurred more strongly than for Steel 2N after a 1000 s holding time. A follow-up analysis then showed that Steel 3N treated at 900 °C revealed a faster coarsening of the carbides than Steel 2N even in the early stage of precipitation, evidencing that Steel 2N exhibited a lower softening resistance at 900 °C. Aims/hypothesis: Islets are thought to be stably present in the adult human pancreas to maintain glucose homeostasis. However, identification of the pancreatic intraepithelial neoplasia (PanIN)–islet complex in mice and the presence of PanIN lesions in adult humans suggest that similar remodelling of islet structure and environment may occur in the human pancreas. To identify islet remodelling in a clinically related setting, we examine human donor pancreases with 3D histology to detect and characterise the human PanIN–islet complex. Methods: Cadaveric donor pancreases (26–65 years old, n = 10) were fixed and sectioned (350 μm) for tissue labelling, clearing and microscopy to detect local islet remodelling for 3D analysis of the microenvironment. The remodelled microenvironment was subsequently examined via microtome-based histology for clinical assessment. Results: In nine pancreases, we identified the unique peri-lobular islet aggregation associated with the PanIN lesion (16 lesion–islet complexes detected; size: 3.18 ± 1.34 mm). Important features of the lesion–islet microenvironment include: (1) formation of intra-islet ducts, (2) acinar atrophy, (3) adipocyte association, (4) inflammation (CD45+), (5) stromal accumulation (α-SMA+), (6) increase in Ki-67 proliferation index but absence of Ki-67+ alpha/beta cells and (7) in-depth and continuous duct–islet cell contacts, forming a cluster. The duct–islet cell cluster and intra-islet ducts suggest likely islet cell neogenesis but not replication. Conclusions/interpretation: We identify local islet remodelling associated with PanIN–islet complex in the adult human pancreas. The tissue remodelling and the evidence of inflammation and stromal accumulation suggest that the PanIN–islet complex is derived from tissue repair after a local injury. [ABSTRACT FROM AUTHOR]

Time-domain reflectometry (TDR) is considered as a passive monitoring technique which reveals multi-functions, such as water level, bridge scour, landslide, and suspended sediment concentration (SSC), based on a single TDR device via multiplexing and related algorithms. The current platform for revealing TDR analysis and interpreted observations, however, is complex to access, thus a coherent data model and format for TDR heterogeneous data exchange is useful and necessary. To enhance the interoperability of TDR information, this research aims at standardizing the TDR data based on the Open Geospatial Consortium (OGC) Sensor Web Enablement (SWE) standards. To be specific, this study proposes a TDR sensor description model and an observation model based on the Sensor Model Language (SensorML) and Observation and Measurement (O&M) standards. In addition, a middleware was developed to translate existing TDR information to a Sensor Observation Service (SOS) web service. Overall, by standardizing TDR data with the OGC SWE open standards, relevant information for disaster management can be effectively and efficiently integrated in an interoperable manner.
