Weltweit leiden immer mehr Frauen an Fertilitätsproblemen und einem unerfüllten Kinderwunsch. Die Granulosazellforschung kann dabei helfen diesen Problemen auf den Grund zu gehen und eine bestmögliche Behandlung zu finden. Die Qualität der Granulosazellen steht in direkter Verbindung mit der Eizellqualität, wodurch die Grundlagenforschung auf diesem Gebiet ein essentielles Mittel zur Bekämpfung der Fertilitätsproblematik darstellt. In dieser Studie soll eine Extraktionsmethode für Granulosazellen aus der Follikelflüssigkeit etabliert werden, durch welche die Zellen einen möglichst hohen Grad an Vitalität und Reinheit aufweisen. Bei erfolgreichen Isolationsprozessen (n=22) nach physikalischen Eigenschaften der in der Follikelflüssigkeit enthaltenen Zellen wurden die Granulosazellen in Kultur gebracht und/oder kryokonserviert. Es wurden verschiedene Methoden der Purifikation getestet: Dichtegradientenzentrifugation mit PureSperm, Percoll, Ficoll oder RBC Lysis Puffer; die Zell-Aggregations-Methode; die Flask Methode; die Haftungs-Methode und die Cell Strainer Methode. Alle Methoden dienen dazu Erythrozyten, Leukozyten, Endothelzellen und Epithelzellen aus der Follikelflüssigkeit zu entfernen. Die Zell Viabilität wurde durch eine Trypanblau Färbung festgestellt und die Anzahl der Zellen wurden mit einer Thoma-Kammer ermittelt.Die besten Ergebnisse konnten mit der Dichtegradientenzentrifugation mit PureSperm und Percoll erzielt werden. Bei der Isolation mit PureSperm konnte eine durchschnittliche Viabilität von 67,77% erreicht werden und bei der Isolation mit Percoll wurde eine durchschnittliche Viabilität von 65,97% dokumentiert. Digitalisierung betrifft jede Generation. In jedem Lebensbereich kommt der Mensch damit in Berührung und dementsprechend hat auch Österreich das Ziel vor Augen, die Gesellschaft in diesem Bereich zu schulen. Mit dem Fach "Digitale Grundbildung" bringt man Informatik und digitale Bildung in die Schulen. Mit DigiFit4All in jedermans Leben. Mit dem DigiFitB4se Kurs verfolgt das Projekt das Ziel, jedem Menschen digitale Kompetenzen über personalisierte offene Onlinekurse zu vermitteln. Die dafür benötigten Materialien, im Kontext auch Lernobjekte genannt, werden auch am Standort Klagenfurt realisiert. Was zeichnet qualitativ hochwertig erstellte Lernobjekte aus? Mit dieser Frage eröffnet sich das Themenfeld dieser Masterarbeit. Mit ALOHA wird ein Qualitätsmodell vorgestellt, dass einerseits auf die Bedürfnisse von DigiFit4All zugeschnitten ist, aber andererseits Lernmaterialien aus allen Kontexten auf ihre Qualität evaluieren lässt. Die 17 Kriterien, auch Skalen genannt, betrachten das Lernobjekt aus didaktischer und technischer Perspektive. Zusätzlich ermöglicht ein Fragebogen in KAUA, einem weiteren Projekt des Instituts für Informatikdidaktik Klagenfurt, die Umsetzbarkeit in der Praxis. Diese Masterarbeit beschreibt die technischen und didaktischen Voraussetzungen, sowie die Entwicklung von ALOHA als Qualitätsmodell und dem zugrundeliegenden Fragebogen als Evaluationsins trument.
