The current study explored whether ALA could prevent testis Pb accumulation and Pb-induced oxidative testicular toxicity in rats. Adult male rats were randomly divided and administered ALA (25 mg/kg/day) and/or Pb (Pb acetate, 20 mg/kg/day) in Control group, ALA group, Pb group, and ALA + Pb group for 4 weeks consecutively. Pb exposure significantly increased Pb accumulation in the testes and blood of Pb-exposed rats compared to the control. Pb caused marked deficits in sperm count, motility along with elevated abnormal sperm cells, while serum testosterone, follicle stimulating hormone (FSH) and leutenizing hormone (LH) levels reduced prominently in comparison to the control. It remarkably depressed the testicular activities of catalase (CAT), glutathione peroxidase (GPx), superoxide dismutase (SOD), as well as levels of interleukin-4 (IL-4) and interleukin-10 (IL-10), whereas malondialdehyde (MDA), tumor necrosis factor-alpha (TNF-α), and interleukin-6 (IL-6) increased significantly compared to the control. The levels of caspase-3, capsase-9, and the PCR gene expression of Bax and Bcl2 were adversely altered with alterations in testis histology. Interestingly, the co-treatment with ALA blocks Pb accumulation in the testis and blood. It inhibited the Pb-induced alterations in reproductive indices and reversed the oxidative stress-mediated inflammatory stress, apoptotic signaling and histopathological abrasions. The findings reveal that ALA could abrogate Pb-induced testicular toxicity through blocking testicular oxidative inflammation and apoptosis in rats.

The use of green nano-amendments is a promising approach for improving soil health and providing sustainable options to reduce root-knot nematodes (RKN) and thus increase yields. Therefore, the purpose of this research was to identify the characteristics of nano-amendments such as nanobiochar (nB), green nanobiochar (GnB), and magnetic nanobiochar (MnB) and their effect on the root-knot nematodes and tomato yield at levels of 3 and 6 mg kg−1 in sandy loam soil. The results showed that the GnB and MnB contain many functional groups (such as O-H, C=C, S-H, H-C=O, C-O, and H–O–H) and minerals (such as magnetite, ferrous sulfate monohydrate, and quartz), and they also had an elevated specific surface area. The application of the investigated soil nano-amendments significantly increased soil organic matter (OM) and microbial biomass carbon (MBC) and decreased the root-knot nematodes, playing a major role in increasing tomato growth. The highest significant values of OM and MBC were found in the soil amended by GnB at 6 mg kg−1, with increases of 84.7% and 71.5% as compared to the control, respectively. GnB6 significantly decreased the number of root galls, the egg mass, and number of nematodes per 250 cm3 soil by 77.67, 88.65, and 74.46%, respectively, compared to the control. Green nanobiochar was more efficient in accelerating the growth and yield components of the tomato plant. The addition of GnB is an effective strategy and an environmentally friendly technology to control plant parasitic nematodes and increase tomato yield. Therefore, the results recommend adding GnB at a rate of 6 mg kg−1 in sandy loam soil.

Recycling waste, such as rice straw and water treatment residuals, is important to reduce harmful effects on the environment and to improve canola yield and soil quality in degraded soils. Nanotechnology for the production of nanomaterials from biochar and water treatment residues will be a future revolution for improving soil quality and increasing canola yield in degraded soil. Therefore, this study aims to identify the properties of some recycled nanomaterials, such as nanobiochar (nB) and nanowater treatment residue (nWTR), and their effect on the biological activity and productivity of canola in degraded soils. The results showed that the nWTR and nB contain many functional groups and minerals, and they also have high negative zeta potential. The addition of the studied soil amendments significantly improved microbial biomass carbon (MBC) and biological activity, which played a major role in increasing canola yield. The highest dehydrogenase (DHA) and catalase (CLA) activity was found in nWTR-treated soil at 50 mg kg−1, with increases of 32.8% and 566.7% compared to the control, respectively. The addition of nB greatly improved the growth of canola plants in the soil. This was evident from the increase in the weight of seeds, the weight of 1000 grains, the number of pods per plant, and the highest increase was for nB added at the rate of 250 mg per kg−1 soil. The addition of 50 mg kg−1 of nWTR gave the best results in seed yield by 150.64% compared to the control. These results indicate that recycled nWTR and nB are some of the best waste recycling treatments, in addition to good soil health, in increasing soil biology and canola yield in degraded soils. In the future, research on recycled nanomaterials should examine the residual effect they have on yield, soil quality, and soil fauna in the long term.

Inflammation prompts cancer development and promotes all stages of tumorigenesis. Calcitriol is a nutraceutical essential regulator for host health benefits. However, the influence of calcitriol on inflammatory mediators involved in cancer cells is not clear. This study aimed to assess the sensitivity of calcitriol alone and combined with capric acid, and identify the possible influence of calcitriol on inflammatory mediators. The colorectal cancer cell line (HCT116) was induced by LPS/TNF-α and the inflammation and metastatic mediators (IL-1β, IL-6, IL-17) were quantified in calcitriol and capric acid supplemented colon cancer cells. The mRNA and protein expression of MMP-2, NF-κB and COX-2 were quantified. The significant reduction in MMP-2 expression was confirmed at combination treatment by zymogram analysis. Our findings demonstrated the anti-inflammatory and anti-metastatic potentials of capric acid and calcitriol in individual exposure in a combination of human colon cancer cell lines (HCT116). These abilities may be due to the inhibition of COX-2 mediators and NF-κB transcription factor and reciprocally regulated MMP-2 and MMP-9 signaling pathways. These findings elucidate the activation of COX-2 and NF-κB via disruption of the cellular outer matrix could be considered a novel molecular target suitable for colorectal cancer therapy. This study confirmed that capric acid activates calcitriol sensitization in colon cancer cells and could be used as a successful supplement for intestinal diseases and colon aberrations.

Human hepatocellular carcinoma (HCC) is the fifth most common cancer and the third leading cause of death across the world. Recent evidence suggests that STAT3 regulates proliferative, survival, metastasis, and angiogenesis genes in HCC. Novel agents that suppress STAT3 activation can be used to prevent or treat HCC. We used a functional proteomics tumor pathway technology platform and multiple HCC cell lines to investigate the effects of acacetin (ACN) on STAT3 activation, protein kinases, phosphatases, products of STAT3-regulated genes, and apoptosis. ACN was found to inhibit STAT3 activation in a dose- and time-dependent manner in HCC cells. Upstream kinases c-Src, Janus-activated kinase 1, and Janus-activated kinase 2 were also inhibited. The ACN inhibition of STAT3 was abolished by vanadate treatment, suggesting the involvement of tyrosine phosphatase activity. ACN was found to suppress the protein expression of genes involved in proliferation, survival, and angiogenesis via STAT3 inhibition. ACN appears to be a novel STAT3 inhibitor and may be a promising therapeutic compound for application in the treatment of HCC and other cancers.

Plants from the family Brassicaceae produce brassinin (BSN), which is an essential indole phytoalexin. BSN can kill certain types of cancer cells. Using hepatocarcinoma (HCC) cells, we examined the molecular mechanisms of BSN. We found that HCC cell growth was suppressed and apoptosis was induced by BSN via the downregulation of the JAK/STAT3 pathway. The cytoplasmic latent transcription factor STAT3, belonging to the STAT family, acted as both a signal transducer and an activator and was linked to tumor progression and decreased survival. BSN incubation caused HCC cells to produce reactive oxygen species (ROS). By activating caspase-9/-3 and PARP cleavage, Bcl-2 was reduced, and apoptosis was increased. BSN inhibited constitutive STAT3, JAK2, and Src phosphorylation. The JAK/STAT signaling cascade was confirmed by siRNA silencing STAT3 in HCC cells. BSN also suppressed apoptosis by Z-Val-Ala-Asp-Fluoromethylketone (Z-VAD-FMK), an apoptotic inhibitor. N-acetylcysteine (NAC) inhibited the production of ROS and diminished BSN-induced apoptosis. Our findings suggested that BSN has potential as a treatment for cancer.

Rutin, a food derived-polyphenolic bioflavonoid, has been acknowledged for several health benefits. This study aims to explore the ameliorative effects of rutin against carbon tetrachloride (CCl4) toxicity in male rats. Adult male rats were given either CCl4 (30% in olive oil, 3 ml/kg b.w. intraperitoneally) alone or in combination with rutin (70 mg/kg intragastrically) twice a week for 4 weeks. Our data showed that rutin mitigated CCl4 hepatorenal damage, as indicated by diagnostic markers (i.e., transaminases, alkaline phosphatase, total bilirubin, total protein, albumin, urea, uric acid and creatinine), and histopathological findings. In addition, CCl4 induced profound elevation of free radical generation and oxidative stress, as evidenced by increasing lipid peroxidation and reducing catalase, superoxide dismutase and glutathione peroxidase activities in liver, kidney and testicular tissues; these effects were suppressed by coexposure with rutin. Moreover, the increase in the levels of serum triglycerides, cholesterol, low-density lipoprotein cholesterol, and very-low-density lipoprotein cholesterol induced by CCl4 was effectively counteracted by rutin. The decrease in the level of high-density lipoprotein cholesterol in the CCl4 group was also counteracted by rutin treatment. Interestingly, the decreased levels of hormonal mediators associated with sperm production, including serum testosterone, luteinizing hormone and follicle-stimulating hormone, and the impaired sperm quality induced by CCl4 were reversed by rutin. Data from the current study clearly demonstrated that rutin supplementation could at least partly overcome CCl4-induced hepatotoxicity, nephrotoxicity and reproductive toxicity by antioxidant and antidyslipidemic effects.

In many sustainable agricultural systems, combining soil ameliorants with deficit irrigation is a viable strategy. However, little is known about how this approach affects soil quality, crop water productivity (CWP), and sugar beet (Beta vulgaris L.) yield in sandy soils. This study investigates the effects of different soil ameliorants―potassium polyacrylate (PPA), polyacrylamide (PAM), and humus (HA)―on the soil chemical properties, water productivity, and sugar beet yield under three irrigation regimes (100%, 80%, and 50% of crop water requirement (CWR). The results demonstrated that applying these amendments signficantly enhances soil pH, cation exchange capacity (CEC), organic matter (OM), and the availability of essential nutrients (N, P, and K). Notably, the combination of PAM and HA under 50% CWR resulted in the highest improvements in sugar beet biomass, increasing shoot and root growth by 73.43% and 71.68%, respectively. It also led to a 97.91% increase in sugar yield and a 4.22% improvement in sugar quality. However, this treatment had the lowest economic benefit, with a negative outcome. In contrast, PAM treatment under 50% CWR produced a 61.49% increase in sugar yield and a 2.44% improvement in sugar quality, ensuring economic viability. Trimetazidine (TMZ) is a promising emerging therapeutic piperazine derivative for renal pathologies. However, the nephroprotective mechanism of TMZ against heavy metal-induced toxicity is unknown. This study, therefore, aimed to explore whether TMZ could mitigate mercury-induced nephrotoxicity in rats. Rats were injected TMZ (3 mg/kg bw) and/or mercury chloride (HgCl2) (4 mg/kg bw) for 4 days (n = 6 rats per group). The blood analysis revealed marked increases in creatinine, urea and uric acid levels in HgCl2 group compared to the control. HgCl2 induced prominent decreases in renal superoxide dismutase (SOD), catalase (CAT), glutathione peroxide (GPx) activities compared to the control followed by marked increases in the levels of malondialdehyde (MDA), interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), caspase-3 and caspase-9. Whereas the renal levels of anti-inflammatory cytokines interleukin-4 (IL-4) and interleukin-10 (IL-10) reduced considerably compared to the control. Contrarily, it was found that in the rats administered TMZ + HgCl2, levels of renal markers, MDA, TNF-α, IL-6 and caspases-3/-9 were prominently reduced compared to the HgCl2 group. The renal SOD, CAT, GPx, IL-4, and IL-10 were markedly elevated along with ameliorated histopathological lesions. Liver is one of the targets of cadmium (Cd) bioaccumulation for hepatic damage and pathologies via oxidative inflammation and apoptosis. The current study explored whether the citrus flavonoid naringenin (NAR) could prevent hepatic accumulation of Cd and Cd hepatotoxicity in a rat model. Rats in group 1 received normal saline; group 2 received NAR (50 mg/kg body weight); group 3 received CdCl2 (5 mg/kg body weight); group 4 received NAR + CdCl2, for four consecutive weeks. Assays related to markers of oxidative stress, inflammation, and apoptosis were carried out using liver homogenate. Blood and liver sample analyses revealed significant elevation of blood and hepatic Cd levels coupled with prominent increases in alkaline phosphatase (ALP), alanine aminotransferase (ALT), and aspartate aminotransferase (AST) activities, whereas the albumin and total protein levels were decreased considerably. Hepatic superoxide dismutase (SOD), catalase (CAT), glutathione peroxide (GPx) activities diminished significantly compared to control followed by marked increases in malondialdehyde (MDA) levels, and dysregulation in caspase and cytokine (TNF-α, IL-6, IL-4, IL-10) levels. However, it was found that in the rats administered NAR + Cd, the levels of Cd, hepatic enzymes, MDA, TNF-α, IL-6, and caspases-3/-9 were prominently reduced compared to the Cd group. The hepatic SOD, CAT, GPx, IL-4, IL-10, albumin, and total protein were markedly elevated along with alleviated hepatic histopathological abrasions. Taken together therefore, NAR is a potential flavonoid for blocking hepatic Cd bioaccumulation and consequent inhibition of Cd-induced oxidative inflammation and apoptotic effects on the liver of rats. Background: Betamethasone treatment is widely used and recommended worldwide. Although, there are no enough studies to look for the risk or potential alterations due to accumulation of this drug before fertilization. In addition, previous studies payed attention to intramuscular injection during pregnancy neglecting the long-term impact of the use of topical solutions. So, the aim of the present study was to determine this side effect. Methods: We used two doses of betamethasone: 0.02 and 0.2 mg/kg b.w. were and at the end of the experimental period (after 3 weeks of delivery), six new-born rabbits of both sexes (1 new born rabbit per mother) were dissected under light anesthesia and heart samples were collected. All the measurements were performed in one assay. Result: The results of this study revealed maternal exposure to betamethasone prior to fertilization, induced oxidative stress, alterations in levels of enzymes. Further, these biochemical alterations were supported by histopathological and ultrastructural observations. So, this study suggested that females, desiring to be pregnant, should stop treatment of psoriasis with betamethasone before fertilization to avoid heart toxicity of their offspring. Diabetes mellitus (DM) provokes reproductive impairments through endocrine disturbance, sperm deficits, and testicular oxidative inflammation. The study investigated the reproductive protective effects of resveratrol (RSV) against testicular oxidative inflammation, sperm/endocrine deficits, and insulin resistance in streptozotocin- (STZ-, 65 mg/kg) induced DM rat model. Male rats were randomly divided into 4 groups (n = 6): control, DM, RSV (150 mg/kg bw, orally), and RSV+DM group (21 days). The nontreated DM rats showed marked decreases in serum insulin, reproductive hormones (T, LH, and FSH), and lipid profile levels compared to control. The homeostatic index of insulin resistance (HOMA-IR) and the quantitative insulin sensitivity check index (QUICKI) were adversely modulated. Sperm count and motility were profoundly decreased, whereas sperm abnormality was significantly increased. The testicular activities of catalase (CAT), glutathione peroxidase (GPx), superoxide dismutase (SOD), and malondialdehyde (MDA) level, along with inflammatory cytokines (TNF-α, IL-6, IL-4, and IL-10) were significantly dysregulated. The DM induced histopathological lesions compared to control rats. Interestingly, the RSV administration to DM rats attenuated the altered reproductive parameters, restored antioxidant mechanism, and anti-inflammatory responses with improved insulin resistance. Background: Lead (Pb) is a heavy metal that represents an environmental toxicant causing serious health problem. Apigenin (APG) is a plant derived flavonoid with several therapeutic activities. So, the aim of the present study was to determine the protective effect of APG against Pb induced toxicity in cerebella of treated rats. Methods: Twenty-four adult male rats (Rattus norvegicus) were used in the present study. They were divided into four groups of six animals per each one as follows: Group I: Rats were daily injected (i.p.) with normal saline as vehicle. Group II: Rats were daily injected (i.p) with lead acetate at a dose of 20 mg/kg b.w. Group III: Rats were daily injected (i.p) with lead acetate at a dose of 20 mg/kg (b.w.) followed by injection (i.p) with APG at dose of 20 mg/kg b.w. Group IV: Rats were daily injected (i.p) with APG at dose of 20 mg/kg b. w. At the end of the four-weeks experimental period, the animals were sacrificed and samples of blood and cerebellar tissue were collected. Result: Lead induced oxidative stress, alterations in levels of studied neurotransmitters, increase in the activities of acetylcholinesterase and increase in the level of the Il-6 and TNF-a and decrease in Il-4 and Il-10 and histological alterations in cerebellum. Background: Deoxynivalenol (DON) is a trichothecene mycotoxin with demonstrated cytotoxicity in several cell lines and animals, primarily owing to inflammation and reactive oxygen species accumulation. Ruscogenin (RGN), a steroidal sapogenin of Radix Ophiopogon japonicus, has significant anti-thrombotic/anti-inflammatory effects. Objective: The aim of this study was to assess the protective role of RGN against DON-induced oxidative stress, which occurs through the nuclear factor erythroid 2-related factor 2 (Nrf2) pathway and is regulated by phosphoinositide 3-kinases/protein kinase B (PI3K/AKT). Methods: The effects were examined using the HepG2 cell line. RGN and DON were suspended in serum-free medium. Cells were seeded onto plates, and then RGN, DON, or both were added over 24 h in triplicates for each group. Results: RGN conferred protection against DON-exhibited cytotoxicity against HepG2 cells. RGN pretreatment downregulated the expression of DON-induced TNF-α and COX-2 and the formation of reactive oxygen species in a dose-dependent manner. RGN upregulated the expression of Nrf2 and its antioxidant proteins as well as mRNA levels of HO-1/NQO-1/HO-1/Nrf2. Similarly, treatment with DON + RGN resulted in upregulation of the pI3K/pAKT signaling pathway in a dose-dependent manner. Finally, RGN was also found to inhibit the DON-induced apoptosis by upregulating the levels of cleaved proteins and downregulating the expression of Bcl2. Result: Maternal exposure to betamethasone, for 2 months before fertilization, induced toxicity in new-born rabbits and decrease the relative organ weight of liver and kidney in new-born rabbits. The biochemical changes involved induction of oxidative, alterations in hepatic and renal parameters. Moreover, structural changes in liver and kidney new-born rabbits were observed. Human hepatocellular carcinoma (HCC) is the fifth most common cancer and the third leading cause of death across the world. Recent evidence suggests that STAT3 regulates proliferative, survival, metastasis, and angiogenesis genes in HCC. Novel agents that suppress STAT3 activation can be used to prevent or treat HCC. We used a functional proteomics tumor pathway technology platform and multiple HCC cell lines to investigate the effects of acacetin (ACN) on STAT3 activation, protein kinases, phosphatases, products of STAT3-regulated genes, and apoptosis. ACN was found to inhibit STAT3 activation in a dose- and time-dependent manner in HCC cells. Upstream kinases c-Src, Janus-activated kinase 1, and Janus-activated kinase 2 were also inhibited. The ACN inhibition of STAT3 was abolished by vanadate treatment, suggesting the involvement of tyrosine phosphatase activity. ACN was found to suppress the protein expression of genes involved in proliferation, survival, and angiogenesis via STAT3 inhibition. Background: Betamethasone is a synthetic glucocorticoid used for treatment of Psoriasis. Until now, the side effect of maternal exposure to topical betamethasone, prior to fertilization, on newborn is not clear. So, the aim of the present study was to determine this side effect. Methods: We used two doses of betamethasone: 0.02 and 0.2 mg/kg b.w. At the end of the experimental period (after 3 weeks of delivery), six new-born rabbits of both sexes (1 newborn rabbit per mother) were dissected under light anesthesia and blood and cerebellum samples were collected. All the measurements were performed in one assay. Result: The results of this study revealed that betamethasone can bioaccumulate in sera of rabbits treated mothers. Maternal exposure to betamethasone, for 2 months before fertilization, induced oxidative stress, alterations in levels of studied neurotransmitters and increase in activities of acetylcholinesterase and caspase-3 enzymes. Furthermore, these biochemical alterations were supported by histopathological observations. So, this study suggested that females; desiring to be pregnant, should stop treatment of psoriasis with betamethasone before fertilization to avoid cerebellar toxicity of their offspring. The current study aims for the use of the solid-state technique as an efficient way for the preparation of zinc oxide nanoparticles (ZnONPs) as an antimicrobial agent with high concentration using sodium alginate as stabilizing agent. ZnONPs were prepared with three different concentrations: ZnONPs-1, ZnONPs-2, and ZnONPs-3 (attributed to the utilized different concentrations of zinc acetate, 1.5, 3 and 4.5 g, respectively). The as-fabricated ZnONPs (ZnONPs-1, ZnONPs-2, and ZnONPs-3) were used for the treatment of cellulosic fabrics as dressing materials for the diabetic wounds. DLS findings illustrated that the as-prepared ZnONPs exhibited average particle size equal to 78, 117, and 144 nm, respectively. The data also showed that all the formulated ZnONPs were formed with good stability (above −30 mv). The topographical images of cellulosic fabrics loaded with ZnONPs that were obtained by SEM confirmed the deposition of nanoparticles onto the surface of cellulosic fabrics with no noticeable agglomeration. The findings also outlined that the treated cellulosic fabrics dressings were proven to have enhanced bactericidal characteristics against the pathogenic microorganisms. The finding of wound contraction for the diabetic rats was measured after 21 days and reached 93.5% after treating the diabetic wound with cotton fabrics containing ZnONPs-2. Ultimately, the generated wound dressing (cellulosic fabrics loaded with ZnONPs) offers considerable promise for treating the wound infections and might be examined as a viable alternative to antibiotics and topical wound treatments. Plants from the family Brassicaceae produce brassinin (BSN), which is an essential indole phytoalexin. BSN can kill certain types of cancer cells. Using hepatocarcinoma (HCC) cells, we examined the molecular mechanisms of BSN. We found that HCC cell growth was suppressed and apoptosis was induced by BSN via the downregulation of the JAK/STAT3 pathway. The cytoplasmic latent transcription factor STAT3, belonging to the STAT family, acted as both a signal transducer and an activator and was linked to tumor progression and decreased survival. BSN incubation caused HCC cells to produce reactive oxygen species (ROS). By activating caspase-9/-3 and PARP cleavage, Bcl-2 was reduced, and apoptosis was increased. BSN inhibited constitutive STAT3, JAK2, and Src phosphorylation. The JAK/STAT signaling cascade was confirmed by siRNA silencing STAT3 in HCC cells. BSN also suppressed apoptosis by Z-Val-Ala-Asp-Fluoromethylketone (Z-VAD-FMK), an apoptotic inhibitor. N-acetylcysteine (NAC) inhibited the production of ROS and diminished BSN-induced apoptosis. The aim of this study was to evaluate the probable protective effect of quercetin (QUE) against cadmium (Cd)-induced sub-chronic toxicity in rats. Adult male rats were given either Cd (as cadmium chloride; 5 mg/kg) alone or in combination with QUE (50 mg/kg) daily for 4 weeks by oral gavage. At the end of the experimental period, Cd accumulation, and selected hematological, thyroid, and reproductive markers were assessed. Results revealed that Cd treatment significantly increased Cd concentrations in blood, thyroid gland, and testicular tissue of rats. Cd also caused a decline in hemoglobin content, hematocrit value, and total erythrocyte and leucocyte counts. Further, significant suppressions in the blood levels of hormones related to thyroid gland function, and male reproductive hormones (i.e., testosterone, luteinizing hormone and follicle-stimulating hormone), were observed in Cd-treated rats compared to the control. In parallel, low sperm count and sperm motility, increased sperm abnormalities, and marked pathology occurred in testis. Combination with QUE recorded amelioration of the deleterious effects of Cd, involving regulation of hematological toxicity and thyroid hormonal levels and subsequently modulation of testicular function. In conclusion, it appears that dietary QUE can rescue from Cd-induced hematological dysfunctions and testicular damage by reversing the hypothyroid state. [ABSTRACT FROM AUTHOR]
