Abstract We studied the effect of dimethyl sulfoxide (DMSO) on the biochemical and physiological parameters of S. cerevisiae yeast cells with varied energy metabolism and antioxidant status. The wild-type cells of varied genetic backgrounds and their isogenic mutants with impaired antioxidant defences (Δsod mutants) or response to environmental stress (ESR) (Δmsn2, Δmsn4 and double Δmsn2msn4 mutants) were used. Short-term exposure to DMSO even at a wide range of concentrations (2–20%) had little effect on the metabolic activity of the yeast cells and the stability of their cell membranes, but induced free radicals production and clearly altered their proliferative activity. Cells of the Δsod1 mutant showed greater sensitivity to DMSO in these conditions. DMSO at concentrations from 4 to 10–14% (depending on the strain and genetic background) activated the ESR programme. The effects of long-term exposure to DMSO were mainly depended on the type of energy metabolism and antioxidant system efficiency. Yeast cells with reduced antioxidant system efficiency and/or aerobic respiration were more susceptible to the toxic effects of DMSO than cells with a wild-type phenotype and respiro-fermentative or fully fermentative metabolism. These studies suggest a key role of stress response programs in both the processes of cell adaptation to small doses of this xenobiotic and the processes related to its toxicity resulting from large doses or chronic exposure to DMSO.

Zinc oxide nanoparticles (ZnONPs), which are produced on a large scale, pose a potential threat to various environments because they can interact with the microbial populations found in them. Bacteria that are widespread in soil, water, and plant material include the Bacillus cereus group, which plays an important role in biodegradation and the nutrient cycle and is a major factor determining ecological balance. This group includes, among others, the foodborne pathogen B. cereus sensu stricto (herein referred to as B. cereus). The aim of this study was a comprehensive assessment of the effects of commercially available ZnONPs on B. cereus. The MIC (minimum inhibitory concentration) for B. cereus was 1.6 mg/mL, and the MBC (minimum bactericidal concentration) was 1.8 mg/mL. Growth of B. cereus was inhibited by a concentration of ZnONPs lower than or equal to MIC50. Concentrations from 0.2 to 0.8 mg/mL inhibited the growth of these bacteria in liquid media, induced symptoms of oxidative stress, and stimulated an environmental stress response in the form of biofilm and endospore formation. In addition, ZnONPs negatively affected the ability of the bacteria to break down the azo dye Evans Blue but enhanced the antimicrobial properties of phenolic compounds. Sublethal concentrations of ZnONPs generally decreased the activity of B. cereus cells, especially in the presence of phenolics, which indicates their potential toxicological impact, but at the same time they induced universal defence responses in these cells, which in the case of potential pathogens can hinder their removal.

Iodine is an essential trace element for humans, and iodine deficiency is a significant health problem. In this study, an improved method for iodine biofortification based on seed germination was established. Solutions of KI (0.15, 0.30, 0.75 and 1.5 mg∙g−1 of seeds) were applied to germinating radish seeds of two cultivars Raphanus sativus L. var. sativus: Warta and Zlata. Compared with the control (seeds treated with water) the iodine content (in the radish sprouts produced by germinating seeds treated with KI were approximately 112.9–2730 times higher. The application KI rates did not adversely affect the biological quality of the radish sprouts. Regarding the biological quality of the iodine-enriched seedlings, we determined their length, dry mater, protein, soluble sugars, chlorophylls, total phenol, ascorbic acid, thiol group content and total antioxidant capacity. The effect of potassium iodide on the selected parameters of their biological quality varied depending on the KI doses and radish cultivars. The results showed that the most appropriate biofortification application rates were 0.15 and 0.30 mg KI per g seeds, because the enriched seedlings had excellent biological quality parameters.

In our investigation, the chemical composition and bioactive potential of leaf buds of raspberry, blackberry, and a raspberry-blackberry hybrid were determined. Antioxidant and antimicrobial properties were tested in water (W), ethanol-water (EW), and glycerol-water (GW) extracts from the buds. These plant organs contain relatively large amounts of minerals, especially Fe. The total antioxidant capacity (TAC) measured by the ABTS and DPPH methods ranged from 2.86 to 12.19 and 6.75 to 24.26 mmol per 100 g fresh weight (FW) of buds, respectively. TAC values were generally higher in the raspberry than in the case of blackberry and raspberry-blackberry hybrid extracts. The antioxidant properties of the extracts were strongly positively correlated with their content of total phenolic (TP). No such relationship was noted for ascorbic acid (AA), whose concentration in all extracts was at a similarly low level. Antioxidant properties determined in vitro were confirmed for the GW extract from raspberry leaf buds in biological test based on the growth parameters of Δsod1 Saccharomyces cerevisiae mutant cells in hypertonic medium. The extracts also exhibited strong antibacterial properties against Staphylococcus aureus and Enterococcus faecalis and weaker against Enterobacter aerogenes. The studied leaf buds could be therefore an unconventional source of minerals, natural antioxidants and antibacterial compounds with potential applications in the food, pharmaceutical, and cosmetics industries.

The study analysed the effect of silver nanoparticles (AgNPs) on seed germination, field emergence and the physiological parameters of seedlings of two bean cultivars, ‘Bali’ and ‘Delfina’, under normal and chill temperatures. AgNPs solutions (0.25, 1.25 and 2.5 mg dm−3) were applied together with the microbial preparation Nitragina (containing Rhizobium leguminosarum bv. phaseoli) on seeds as a short-term pre-sowing treatment. Low concentrations of AgNPs (0.25, 1.25 mg dm−3) had an immediate beneficial effect, resulting in fast and uniform germination in laboratory and field conditions, as well as a positive effect in the later stages of seedling development, manifested as an increase in the average seedling height, fresh and dry weight and net photosynthesis. Particularly, favourable effects were noted in suboptimal temperature conditions, suggesting that AgNPs activate plant mechanisms of tolerance to environmental stress. The highest concentration tested of AgNPs was not particularly effective for the plants but had a strong antimicrobial effect, which was beneficial in period of seed germination, but at the later stage of plant development was unfavourable probably due to disruption of symbiosis between the bean seedlings and rhizobia.

Iodine deficiency disorders are one of the serious worldwide public health problem in the world. The need to search for alternative methods of iodine supplementa-tion results from the recommendation of the World Heath Organization and aims to sig-nificantly reduce iodine malnutrition in humans diet. Iodine is not included among essen-tials nutrients for plants, but the plants are able to accumulate it. Seedlings biofortified with iodine can become an alternative source of this element for humans. The aim of the study was to attempt to obtain iodine-fortified lettuce and radish seedlings and to deter-mine the effect of the level of iodine applied in the form of potassium iodide on their bio-logical quality. The following levels of KI were used: 0 (control), 0.075, 0.15, 0.0375, 0.75 and 1.5 mg per Petri dishes. The effect of potassium iodide on the selected parame-ters of their biological quality varied depending on the KI doses and species of plant. The seedlings grown in the presence of KI had a higher iodine content. The results showed that the most appropriate biofortification application rates were 0.075 and 0.15 mg be-cause the enriched seedlings had biological quality parameters similar to the control. Sta-tistically significant differences in the parameters characterizing seedling quality were noted most often in the case of the highest amounts of KI (0.375–1.5 mg). These KI con-centrations reduced seedling’s lenght in radish and lettuce seedling but increased dry weight only in lettuce. A significant increase in ascorbic acid concentration only in the lettuce seedlings was obtained. In comparison with the control, no significant differences in the content of biomass and chlorophyll content were noted in the biofortified seedlings. Thiol group content was decreased in both radish and lettuce, but the antioxidant activity measured by DPPH method only in lettuce seedling extracts.

Phenolic compounds are important plant metabolites associated with the plant's defence reactions in conditions of biotic and abiotic stress. Heterotrophic organisms are not capable of synthesizing phenolic compounds, but absorb them with foods of plant origin. Many of them exhibit antioxidant activity in chemical and biological assays. This study examined whether these compounds are able to protect cells of a heterotrophic organism from the effects of severe environmental stress. A wild-type strain of Saccharomyces cerevisiae yeast was used in the experiments. Yeast cells that were pre-incubated in conditions of severe thermal, osmotic or oxidative stress and then plated on a medium enriched with benzoic acid and its derivatives had a similar survival rate to the cells growing on a medium with no supplements, while in the presence of cinnamic acid derivatives and coumarin a reduction was observed in their survival. In the presence of the synthetic monophenol BHT, two different reactions were noted - a decrease in survival after 10 min of severe heat shock, and similar survival to that of the cells growing on the non-supplemented medium after 5 min of heat shock and the other types of stress. The results obtained suggest that monophenols used after severe environmental stress are not capable of eliminating the detrimental effects of these factors. [ABSTRACT FROM AUTHOR]
