Abstract
- In dieser Arbeit konnte zum ersten Mal mithilfe des CBMN-Assays die genaue absorbierte Dosis in den Peaks und Valleys der Mikrostrahlen bestimmt werden. Die biologische Dosimetrie mit Mikrokernen und Brücken konnte die physikalische Dosimetrie validieren. Weiterhin wurde gezeigt, dass rekonstruiertes Hautgewebe gut geeignet ist, um die örtliche Auflösung von DNA-Schäden nach Mikrostrahltherapie zu untersuchen. Hier konnte in den Valleys im Vergleich zu homogener Bestrahlung eine höhere Focianzahl beobachtet werden, was auf mögliche Bystander Effekte hinweisen könnte.<br />This is the first study precisely determining the absorbed doses in the peaks and valleys of microbeams using the CBMN-Assay. The biological dosimetry with micronuclei and bridges agrees with the physical dose determined with Gafchrom film and could therefore validate physical dosimetry. The study clearly demonstrated that reconstructed skin tissue is well suited to study spatial resolution of DNA damage after microbeam therapy. Here, a higher 53BP1 foci number could be observed in the valleys compared to homogeneous irradiation, which could indicate possible bystander effects.
