Total synthesis of 10-deoxymethynolide, the aglycon of the macrolide antiobitic 10-deoxymethymycin

A study was conducted to characterize the preparation of 10-deoxymethynolide, the aglycon of 10-deoxymethymycin. An enantioselective aldol reaction of a chiral beta-tosyloxy aldehyde supported the synthesis delta-lactone. The construction of the dodecanolide ring system was evaluated using a carbon-carbon bond forming approach. Results indicated that the synthesis of 10-deoxymethynolide supported an expeditious process of delta-lactone.
