Abstract
- A sensitive method for detection of alanine formed in the SufS reaction was developed to identify the participation of Fe-S cluster scaffold SufU as a substrate in the cysteine desulfurase gene sufS catalytic mechanism in Gram-positive bacteria, such as Bacillus subtilis. Steady-state kinetic analysis of alanine formation provided evidence for the double-displacement mechanism (ping-pong) of the cysteine:SufU sulfurtransferase reaction catalyzed by SufS.
