Misincorporation of 2'-deoxyoxanosine 5'-triphosphate by DNA polymerases and its implication for mutagenesis

Abstract
- A study was conducted to characterize the formation of 2-deoxyoxanosine 5'-triphosphate (dOTP) from 2'-deoxyguanosine 5'-triphosphate (dGTP) by HNO2 treatment. The study also analyzed the misincorporation of dOTP by DNA polymerases during the replication of M13mp18 and oligodeoxynucleotide templates. Results indicated that the nitrous acid treatment of dGTP in solution supported the synthesis of dOTP and 2'-deoxyxanthosine 5'-triphosphate.
