Proofing of photolithographic DNA synthesis with 3',5'-dimethoxybenzoinyloxycarbonyl-protected deoxynucleoside phosphoramidites

Abstract

A study was conducted on the photochemical solid-phase phosphoramidite DNA synthesis technique. The findings indicate that the coupling efficiency of these amidites in the synthesis of homopolymers was in the range of 80-97%, with purines having lower efficiency than pyrimidines. Moreover, the results suggest that light-directed in situ synthesis of short oligonucleotides will be an effective approach in preparing hybridization surfaces for detection of complementary sequences.
