Abstract
- A kit method devised for purification of bacterial artificial chromosomes (BACs) could be adapted for effectively preparing individual plasmids up to 220 kb from wild gram-negative and gram-positive bacteria. A protocol is described for using solid-phase reversible immobilization (SPRI) for rapid, efficient and inexpensive isolation of sequencing-quality DNA of individual large, low-copy number plasmids from gram-negative and gram-positive bacterial strains.
