Abstract

An interaction Western blotting method facilitates the analysis of interactions between hydrophobic repeated domains (HBD) and enzymatic subunits of the cellulosome. Vector construct of a hybrid protein prepared from Escherichia coli maltose-binding protein renders the overexpression of HBDs of cellulose-binding protein (CbpA) from Clostridium cellulovorans. Immunoblotting interactive method exhibits specific interactions of HBD with endoglucanases, EngB and EngD, and identifies HBD as the endoglucanase binding site of CbpA.
