The hydrophobic repeated domain of the Clostridium cellulovorans cellulose-binding protein (CbpA) has specific interactions with endoglucanases

An interaction Western blotting method facilitates the analysis of interactions between hydrophobic repeated domains (HBD) and enzymatic subunits of the cellulosome. Vector construct of a hybrid protein prepared from Escherichia coli maltose-binding protein renders the overexpression of HBDs of cellulose-binding protein (CbpA) from Clostridium cellulovorans. Immunoblotting interactive method exhibits specific interactions of HBD with endoglucanases, EngB and EngD, and identifies HBD as the endoglucanase binding site of CbpA.
