Analysis of the Bradyrhizobium japonicum hemH gene and its expression in Escherichia coli

Abstract
- Complementation analysis indicates that the Bradyrhizobium japonicum hemH gene is essential to free mutant strains I110ek4 and I110bk2 with respect to hemin auxotrophy, protoporphyrin accumulation and the lack in ferrochelatase activity. Its expression in Escherichia coli give 39-kDa proteins. The overexpressed protein indicates ferrochelatase activity, implying that the hemH gene encodes ferrochelatase. The B. japonicum hemH gene complements the Escherichia coli mutant by exhibiting hemin prototrophy, loss of protoporphyrin accumulation and expression of ferrochelatase activity.
