Toca-1 mediates Cdc42-dependent actin nucleation by activating the N-WASP-WIP complex

Abstract

The biochemical purification of Toca-1 (transducer of Cdc42-dependent actin assembly) as an essential component of the Cdc42 pathway is described. It is found that in this model, activated Cdc42 interacts with both Toca-1 and the N-WASP-WIP complex, and these interactions lead to the activation of N-WASP, which in turn stimulates actin nucleation through the Arp2/3 complex.
