Abstract
- A saturated linkage map of Theobroma cacao has been constructed. A test cross population of 182 individuals has been produced by a cross between two heterozygous genotypes: UPA 402, an Upper Amazon Forastero, and UF 676 a Trinitario genotype. Several kind of markers have been used for mapping and Join Map Software were used for the mapping analyses. This map comprised 357 markers: RFLP: a genomic and a cDNA libraries have been constructed and 170 RFLP markers were mapped. RAPD: primers from Operon tech. were used for this study and provided 30 loci. Isozymes: 5 enzymes were mapped. Microsatellites: 15 microsatellite markers were mapped. AFLP : about 137 AFLP markers were localized in the map. This map length is 872 cM, it comprises 10 linkage groups corresponding to the ten putative chromosome number of T. cacao. The mean distance between two markers is 2.4 cM. (Texte intégral)
