Phenetic relationship and identification of subtribe Oncidiinae genotypes by random amplified polymorphic DNA (RAPD) markers

Abstract

Molecular markers among 24 accessions of subtribe Oncidiinae genotypes were investigated based on random amplified polymorphic DNA (RAPD) analysis. Eighty decamer primers were screened, and 14 primers producing clear and reproducible DNA patterns were selected. In total, 263 bands were scored, of which 257 revealed polymorphism. Phenetic similarity based on the percentage of band sharing was estimated, ranging from 0.25 to 0.71 among the 24 accessions. Cluster analysis based on molecular data of band distribution in all the samples showed six major clusters and one individual not belonging to any of the six clusters. In addition, the genus Miltonia is segregated from other genera among the Oncidiinae in this study. DNA markers at molecular level provide an appropriate tool for identifying Oncidiinae genotypes.
