Culture of 293 cells in different culture systems: Cell growth and recombinant adenovirus production

Abstract

Growth of 293 cells (human embryonic kidney) was compared in various cell culture systems including static flasks, cell aggregates and a variety of porous microcarriers. The best results were achieved with Fibra-Cel carriers and cell aggregates (1.1–1.4 × 106 cells/ml). Virus production was compared using a recombinant replication-deficient adenovirus as a model. Virus yields of lysates from cells grown on Fibra-Cel carriers, as cell aggregates and in static flasks were comparable (1.7–1.9 × 108 pfu/ml).
