ChemInform Abstract: An Enantioselective Synthesis of the Resorcylic Acid Lactone L-783,277 via Addition of an Acetylide Anion to a Tethered Weinreb Amide

The resorcylic acid lactone L-783,277 (V), a potent and irreversible inhibitor of nitrogen-activated protein kinase, is prepared via macrocyclization of alkyne (II) followed by a Lindlar-type hydrogenation and a subsequent deprotection of compound (IV) with BCl3 as key steps.
