Abstract

A multilayer dsDNA-electrochemical biosensor for the in situ electrochemical evaluation of anticancer antibody rituximab (RTX) interaction with double-stranded DNA (dsDNA), with differential pulse voltammetry was used. The decrease of the dsDNA oxidation peaks with incubation time was observed and was confirmed that RTX binds causing dsDNA condensation, and no evidence for DNA base oxidative damage was detected electrochemically. Nondenaturing agarose gel-electrophoresis of RTX-dsDNA samples confirmed the occurrence of condensation in the dsDNA structure.
