Abstract

Chromatin dynamic during normal cell development and differentiation plays a pivotal role in cell functioning. Cellular processes such as DNA replication, transcription, and translation depend on chromatin structure. Therefore modifications of DNA and nuclear proteins—histones—play a key role in these processes and consequently in cell functioning and regulation. Chapter 3 represents effects of a large scale of DNMTi, HMTi and HDACi alone in various inter-combinations and/or combinations with granulocytic differentiation inducer all-trans retinoic acid. Epigenetic reprogramming events including reorganization of DNA and histone modifications signatures, alterations in transcription factors binding to transcription machinery activity, protein expression level changes in in vitro (leukemia cell lines), and in vivo (leukemic xenografts) model systems are presented.
