Decrease in toxicity and mutagenicity of cyclophosphane in rats under the action of tularemia vaccine

Abstract

The experiments were conducted on 186 Wistar rats with an average weight of 140 g. We used CP produced by the Saransk Medical Preparations Factory. The maximum tolerable dose of CP in the case of a single intraperitoneal injection is i00 mg/kg for rats. Considering that the drug exhibits a highly cummulative toxic effect, and on the basis of the literature data [i], to produce the maximum toxic effect CP was injected in 25 mg/kg doses intraperitoneally for four days (total dose i00 mg/kg). The toxicity of CP was assessed according to changes in the following parameters: body weight, relative weight of the spleen (RWS the ratio of the weight of the spleen in milligrams to the body weight in grams), number of lymphocytes, and lifetime of the animals, over a month after administration of the drug. The influence of CP on the intensity of antitularemic immunity was determined according to antibody formation by the agglutination reaction, which is extremely specific in tularemia.
