Alternativni domaćini uvijenosti lista vinove loze pridruženog virusa 3 i A-virusa vinove loze

Abstract

Uvijenosti lista vinove loze pridruženi virus 3 (GLRaV) i A-virus vinove loze (GVA) smatraju se ekonomski važnim virusima vinove loze. Europska vinova loza (Vitis vinifera L.) i američke autohtone vrste te njihovi hibridi smatraju se njihovim glavnim domaćinima, ali su također zabilježeni prijenosi na neke zeljaste test biljke. Pored prijenosa sadnim materijalom, oba virusa su prenosiva štitastim ušima. Pokusi u laboratorijskim uvjetima, pomoću lozine štitaste uši (Planococcus ficus), pokazali su upješan prijenos GVA sa vinove loze na dva česta korova u hrvatskim vinogradima: mračnjak (Abuthilon theophrasti) i oštrodlakavi šćir (Amaranthus retroflexus) te na dvije test biljke: Chenopodium murale i Nicotiana benthamiana; te prijenos GLRaV-3 na korove: bijela loboda (Chenopodium album), sitnocvjetna konica (Galinsoga parviflora) i test biljke Nicotiana benthamiana i Chenopodium murale. Također, utvrđena je i mogućnost povratnog prijenosa na lozu i to sa oštrodlakavog šćira za GVA te sitnocvjetne konice i Chenopodium murale za GLRaV-3. Uspješnost prijenosa potvrđena je metodom ELISA i/ili lančanom reakcijom polimeraze nakon obrnutog prepisivanja (RT-PCR). Navedena otkrića mijenjaju dosadašnja saznanja o epidemiologiji ova dva virusa vinove loze. Grapevine leafroll-associated virus 3 (GLRaV-3) and Grapevine virus A (GVA) are considered economically important viruses of grapevine. The European grapevine (Vitis vinifera L.) and American grapevine species and their hybrids are considered to be their main hosts, but also transfer to some test plants was recorded. In addition to the transmission by infected planting material, both are transmitted by mealybugs. Experiments in laboratory conditions, using vine mealybug (Planococcus ficus), showed successful transmision of GVA from grapevine to two common weeds in Croatian vineyards: velvetleaf (Abuthilon theophrasti) and redroot pigweed (Amaranthus retroflexus) and two test plants: Chenopodium murale and Nicotiana benthamiana; and transfer of GLRaV-3 to common weeds: common lambsquarters (Chenopodium album), and smallflower galinsoga (Galinsoga parviflora) and test plants Nicotiana benthamiana and Chenopodium murale. Also, the possibility of retransmission to the grapevine was determined from redroot pigweed for the GVA and smallflower galinsoga and Chenopodium murale for GLRaV-3. The success of transmission was confirmed by the ELISA and/or polymerase chain reaction after reverse transcription (RT-PCR). These findings give additional knowledge to the epidemiology of these two grapevine viruses.
