Abstract

ABC transporteri su membranski proteini koji koriste energiju ATP-a za prijenos različitih molekula preko stanične membrane. Prisutni su kod svih organizama s određenim razlikama u načinu djelovanja. Istraživanjem molekularnih mehanizama kod malignih oboljenja, otkriveno je da pojedini ABC proteini imaju potencijalnu ulogu u razvoju rezistencije tumorskih stanica na lijekove. To su: P-glikoprotein, MRP1 i BCRP. Danas, sve veći broj ljudi obolijeva od malignih bolesti i liječenje često biva neuspješno. Jedan od razloga neuspješnog liječenja su upravo i ABC transporteri koji mogu utjecati na apsorpciju, distribuciju, metabolizam, ekskreciju i toksičnost lijeka. Smatra se da bi razvoj načina inhibicije njihovog djelovanja značilo prekretnicu u kliničkom liječenju malignih bolesti.<br />ABC transporters are membrane proteins that use the energy of ATP to transfer various molecules across the cell membrane. They are present in all organisms with certain differences in the mode of action. By investigating the molecular mechanisms in malignant diseases, it has been discovered that certain ABC proteins have a potential role in the development of drug resistance of tumor cells. These are: P-glycoprotein, MRP1 and BCRP. Today, an increasing number of people are suffering from malignant diseases and treatment is often unsuccessful. One of the reasons for unsuccessful treatment are ABC transporters, which can affect the absorption, distribution, metabolism, excretion and toxicity of the drug. It is a general opinion that the development their inhibitors would mean a turning point in the clinical treatment of malignant diseases.
