Abstract

Researchers from F. Hoffmann-La Roche Ltd. in Basel, Switzerland have conducted a study on high concentration formulations of biologics for subcutaneous administration. They used a reversible protein complex (RPC) approach and a novel in vitro tool called the Subcutaneous Injection Site Simulator (Scissor) to evaluate the fate and behavior of these formulations. The Scissor simulates the changes that a biological drug could encounter as it transitions from a drug product formulation to the hypodermis following subcutaneous injection. The study found that the Scissor can provide a suitable tool to predict the stability of biological drugs in the subcutaneous environment, and in vitro drug release data showed sustained and controlled release of RPC formulations.
