Abstract

A new study conducted by researchers at Johns Hopkins University School of Medicine explores the resistance of pancreatic adenocarcinoma (PDAC) to ferroptosis, an iron-dependent form of cell death. The study found that PDAC exhibits relative resistance to ferroptosis compared to other tumor types, and this resistance is mediated by the hypoxia-inducible transcription factor HIF-2. The researchers identified several HIF-2 target genes responsible for coordinating ferroptosis resistance mechanisms, including genes involved in cysteine import and biosynthesis, as well as mitophagy regulation. The findings suggest that targeting HIF-2 in combination with pro-ferroptotic agents could be a potential therapeutic strategy for pancreatic cancer. [Extracted from the article]
