Abstract
- A recent preprint abstract discusses a study that aims to understand the spatiotemporal dynamics of unfolded protein responses (UPR) in human cells. The study introduces a mass spectrometry-based proteomics strategy called Simultaneous Proteome Localization and Turnover (SPLAT) to measure changes in protein turnover and subcellular distribution during UPR. The researchers found that protein turnover kinetics during UPR varied across different subcellular localizations, with an overall slowdown but an acceleration in endoplasmic reticulum and Golgi proteins involved in stress response. Additionally, the study identified disruptions in proteostasis in sarcomeric proteins as a potential mechanism of carfilzomib-mediated cardiotoxicity.
