Abstract

The potential of anaflatoxin B<subscript>1</subscript> (AnAFB<subscript>1</subscript>) conjugated to keyhole limpet hemocyanin (KLH) as a vaccine (AnAFB<subscript>1</subscript>-KLH) in controlling the carry over of the aflatoxin B<subscript>1</subscript> (AFB<subscript>1</subscript>) metabolite aflatoxin M1 (AFM<subscript>1</subscript>) in cow milk is reported. AFB<subscript>1</subscript> is the most carcinogenic compound in food and foodstuffs amongst aflatoxins (AFs). AnAFB<subscript>1</subscript> is AFB<subscript>1</subscript> chemically modified as AFB<subscript>1</subscript>-1(O-carboxymethyl) oxime. In comparison to AFB<subscript>1</subscript>, AnAFB<subscript>1</subscript> has proven to be non-toxic in vitro to human hepatocarcinoma cells and non mutagenic to Salmonella typhimurium strains. AnAFB<subscript>1</subscript>-KLH was used for immunization of cows proving to induce a long lasting titer of anti-AFB<subscript>1</subscript> IgG antibodies (Abs) which were cross reactive with AFB<subscript>1</subscript>, AFG<subscript>1</subscript>, and AFG<subscript>2</subscript>. The elicited anti-AFB<subscript>1</subscript> Abs were able to hinder the secretion of AFM<subscript>1</subscript> into the milk of cows continuously fed with AFB<subscript>1</subscript>. Vaccination of lactating animals with conjugated AnAFB<subscript>1</subscript> may represent a solution to the public hazard constituted by milk and cheese contaminated with AFs.
