Monolayer cell cultures of embryonic chick skeletal muscle.

A procedure is described for culturing embryonic chick skeletal muscle in monolayer. A detailed description is provided for the dissection, dissociation, preparation of single cell suspension, and planting of cultures. Dissociation is achieved by mechanical rather than enzymatic process. The preparation of embryo extract, growth medium, and collagen for coating tissue culture dishes is also detailed. The procedure includes the conditions necessary for incubation of cultures at two different levels of carbon dioxide and for avoiding the use of antibiotics. [ABSTRACT FROM AUTHOR]
