Abstract

The article presents a study evaluating the novel lacl mutation (C354T), a transgenic mouse mutation detection system that aims to allow the assessment of in vivo mutation frequency and pattern in individual tissues. In an assay of spontaneous mutations, there was a reappearance of C354T in six of seven mutants with a second mutation. It is observed that the frequency of spontaneous doublets was advanced by 2.7-fold and there was greater average spacing between mutations in the doublets. Furthermore, the study reveals the coherence of C354T as a hitch-hiker mutation in one of 38 mutation targets in the Big Blue mouse genome with germline mutation. Results suggest the essence of determining all mutations and analyzing new mutations in murine mutations.
