Stimulating the synthesis of phenolics and polyacetylenes in Lobelia chinensis Lour. plantlets using various bioreactor culture systems: a comparison of parameter effects.

Abstract
- Lobelia chinensis Lour. is an important medicinal plant used in Chinese traditional medicine. Plantlet cultures of L. chinensis were established in balloon-type airlift bioreactors with the aim of producing L. chinensis raw material. The generation of biomass and bioactive chemicals was investigated using a variety of culture techniques, including the continuous immersion bioreactor (CIB), temporary immersion bioreactor (TIB), and temporary immersion with net (TIB-N). Of the varied culture methods tested CIB system found suitable for the production biomass, phenolics and polyacetylenes. CIB cultures were also used to examine the effects of a number of other parameters, such as the salt strength of Murashige and Skoog (MS) medium, concentration of carbohydrates, aeration volume, and inoculum density. It was discovered that CIB cultures supplemented with full strength MS medium, 3% sucrose, 0.1 vvm of aeration volume, and 3 g L<superscript>− 1</superscript> inoculum were appropriate for producing the highest amount of biomass and bioactive compounds. Higher concentrations of phenolics, including catechin (119.49 µg g<superscript>− 1</superscript> DW), phloretic acid (199.40 µg g<superscript>− 1</superscript> DW), salicylic acid (105.27 µg g<superscript>− 1</superscript> DW), myricetin (167.71 µg g<superscript>− 1</superscript> DW), naringin (88.52 µg g<superscript>− 1</superscript> DW), coumaric acid (76.63 µg g<superscript>− 1</superscript> DW), linarin (103.37 µg g<superscript>− 1</superscript> DW), and chlorogenic acid (46.24 µg g<superscript>− 1</superscript> DW) were all found in plantlets cultured in CIB cultures under optimal conditions. Additionally, plantlets cultivated in optimal CIB cultures had the highest quantities of lobetyolinin (70.85 µg g<superscript>− 1</superscript> DW) and lobetyolin (471.25 µg g<superscript>− 1</superscript> DW).Key message: Lobelia chinensis plantlet biomass was generated in continuous immersion bioreactors, and varied variables influencing polyacetylenes, phenolics, were identified.
