Comprehensive Analysis of Non-coding RNAs Expression Profile and miRNA- circRNA-gene Co-expression Network in Developing Uterine Carcinosarcoma.

Abstract
- Objectives: Uterine carcinosarcoma (UCS), often presenting at the advanced stage, is a rare, aggressive form of uterine cancer. We conduct a prospective cohort study by systematically study whole transcriptome sequencing data of uterine carcinosarcoma to gain further insights into the transcriptomic landscape and molecular mechanism of UCS. Methods: RNAs were extracted from tissue samples from five UCS patients and three corresponding non-tumorous tissues adjacent, followed by Illumina sequencing technology for high-throughput sequencing. A functional clustering analysis of differentially expressed genes was subsequently conducted. Results: We identified 1650 upregulated and 1753 downregulated circRNAs, 1632 upregulated and 460 downregulated lncRNAs. The differentially expressed genes (DEGs) mainly involve in IL-7 receptor-based signaling, MAPK signaling pathway, PD-L1 expression and PD-L1 checkpoint pathway and so on. An miRNA-circRNA-gene co-expression network reveals the top five correlated genes, miRNAs and circRNAs in developing UCS. Conclusion: In conclusion, we reveals the non-coding RNAs expression profile and a novel miRNA-circRNA-gene co-expression network shows correlated RNAs and genes in developing UCS. These profiles and correlated RNAs and genes may be potential pathways and targets in the diagnosis and treatment in developing UCS. [ABSTRACT FROM AUTHOR]
