Liquid semen quality of Donggala bulls treated with CEP-2 and egg yolk combination and stored at 5°C.

Abstract

A study was carried out to find out effects of CEP-2 and egg yolk combination as a diluent on liquid semen quality of Donggala bulls stored at 5°C. The study was designed with a 4 x 5 completely randomized design with four diluent combinations as treatments and five semen collection times as replicates. Fresh semen was obtained from three mature selected Donggala bulls. The treatments were P<subscript>0</subscript>C = CEP-2 + no egg yolk, P<subscript>1</subscript>C = CEP-2 diluent + 5% egg yolk, P<subscript>2</subscript>C = CEP-2 diluent + 10% egg yolk and P<subscript>3</subscript>C=CEP-2 diluent + 15% yolk egg. The quality of liquid semen was observed for spermatozoa motility, viability and abnormalities every 24 hours after dilution until the motility was less than 40%. Results indicated that the average motility of spermatozoa after 3 d of storage for P<subscript>0</subscript>C (20.9±3.8%) was significantly lower (P<0.01) than the other treatments, for which the treatments did not differ (P>0.05). The motility for P<subscript>1</subscript>C, P<subscript>2</subscript>C and P<subscript>3</subscript>C was 44.3±8.1%, 46.9±7.2% and 43.9±8.0%, respectively. The viability of spermatozoa after 3 d of storage for P0C (28.0±6.7%) was also significantly lower (P<0.01) than the other treatments, for which the treatment did not differ significantly (P>0.05). The average viability for P1C, P<subscript>2</subscript>C and P<subscript>3</subscript>C was 49.3±6.6%, 51.0±8.5% and 49.1±6.7%, respectively. The abnormality of spermatozoa after 3 d of storage for P<subscript>0</subscript>C (38.4±3.4%) was significantly higher (P<0.01) than the other treatments, for which the treatment did not differ significantly (P>0.05). The average abnormality for P<subscript>1</subscript>C, P<subscript>2</subscript>C and P<subscript>3</subscript>C was 26.9±3.1%, 28.7±2.1%, 29.6±2.2%, respectively. It can be concluded that the use of egg yolks up to 15% in CEP-2 diluent can maintain the motility of spermatozoa in liquid semen of Donggala bulls above 40% for up to 3 d of storage at 5°C.
