Primer screening based on ISSR and RAPD markers for amplification of Katokkon pepper in Tana Toraja and surrounding district, Indonesia.

Abstract
- Katokkon pepper is a type of pepper that is most interested in people because it has a distinctive taste and aroma, especially in Toraja. The accuracy of the genetic diversity of a plant species is largely determined by the appropriate primer for that plant species. The aim of this study was to determine the suitable annealing temperature for amplification of katokkon pepper DNA using ISSR and RAPD primers. Primers that produce clear polymorphic bands will be used as the basis for further analysis of katokkon genetic diversity. Twelve katokkon peppers were collected from three districts in Sulawesi, i.e., Tana Toraja, Enrekang, and Mamasa. Leaves from each individual were then used as DNA sources. Samples would be analyzed in the ISSR and RAPD primer screening process because this research is the preliminary study for genetic diversity analysis on Katokkon pepper. Selection of primers from ten ISSR primers and five RAPD primers showed that all primers were amplified. Clear and polymorphic bands were formed by primer-1, primer-2, primer-3, primer-4, primer-5, primer-7, primer-9, and primer-10 for ISSR primers. RAPD primers were OPA-05, OPG-06, OPAE-11, OPG-19, and OPK-20. Primers are suitable to be used for determining the genetic diversity of Katokkon in further analyses. [ABSTRACT FROM AUTHOR]
