Abstract
- Researchers at Shanghai Jiao Tong University in China have made advancements in point-of-care testing by combining programmable nucleases with isothermal amplification. The researchers developed a temperature control solution using thermotolerant Pyrococcus furiosus Ago (Pf Ago) beads, which prevented interference with loop-mediated isothermal amplification and accurately identified amplicons. The system demonstrated high sensitivity and specificity, with no cross-reactivity with other pathogens. This research presents an effective method for combining amplification and cleavage, improving the clinical applicability of diagnostic techniques dependent on programmable nucleases.
