Abstract

This article discusses a new method and application of PacBio sequencing for low copy and difficulty preparation plasmids. The Single Molecule Real Time (SMRT) system developed by Pacific Biosciences is used for sequencing and utilizes the SMRT chip as the sequencing carrier. The article explains the challenges of using PacBio sequencing for low-copy plasmids and proposes a solution using Phi29 polymerase for multiple substitution amplification. Additionally, the study establishes a PacBio sequencing method for bacterial fluids without the need for plasmid extraction steps, reducing time and cost.
