Evaluating the Potential Role of Mir-320b and C-Myc in Recurrent Pregnancy Loss.

Abstract

Recurrent pregnancy loss (RPL) is a major life event and a common pregnancy complication, which defined as losses two or more pregnancies before 20 weeks of gestation. It affects approximately 1%-2% of reproductive women. To evaluate the potential role of circulating microRNAs (miRNAs) as a non-invasive biomarker in maternal blood and focus on the similarity between tumour invasion and embryogenesis as phenomena by c-Myc. This case control study comprised 50 women with RPL (25 primary RPL and 25 secondary RPL) from two infertility clinics: Infertility Centre, Babylon Teaching Hospital, Babylon province, and Teba Centre, Babylon province, Iraq. And 50 control women with no history of RPL and at least one child birth. Both groups are matched in age and BMI. Blood sampling was done for all and protein level of c-Myc measured by ELISA Kit and miRNA extraction and analysis done according to the manufacturer recommendations. The results presented in this study showed a significant difference in miR-320b Ct value between RPL patient and control (p 0.00) and there is a significant difference in c-Myc level between patient and control (p 0.00). A negative correlation found between miR-320b and c-Myc (p 0.01, r=-0.27). The present study showed that miR-320b could be regarded as a novel biomarker and play a valuable role in RPL pathogenesis along with c-Myc which affect cell proliferation and invasion. However, this binding protein transcription factor is a part of a coordinated network of various set of partners which are needing for further studies in future.
