ISOLATION AND STRUCTURAL ELUCIDATION OF AN TERPENOID COMPOUND FROM ALLIUM SATIVUM AND THEIR NEPHROPROTECTIVE ACTIVITY AGAINST LPS.

Abstract
- Allium sativum contain various medicinal properties like immunomodulatory, antitumorigenetic, antiatherosclerotic and antimicrobial. The endotoxin lipopolysaccharide (LPS) is a dominant component of the outer membranes of Gram-negative bacteria, which is largely responsible for toxic inflammatory reactions. The main objective of this study is isolation and structural elucidation of bioactive compound from Allium sativum and estimates its possible nephroprotective role in LPS intoxicated mice. Fresh bulbs of Allium sativum were collected and identified by a taxonomist. Bulbs were powdered and extracted by soxhlet 85% hydroethanol solvent. The extract was subjected to different chromatography techniques to isolate a novel bioactive compound whose structure was elucidated with the help of FTIR, NMR and LC-MS and determine the effect of hydroethanol extract of Allium sativum in LPS challenged mice. The structure of the isolated compound was elucidated as Terpenoid and its IUPAC name is Isopropyl 2-(3-acetyl-5-(3,3-dimethylbut-1-en-2-yl)phenyl)-4-isopropyl-3-methyleneheptanoate. Allium sativum significantly attenuated the LPS-induced renal toxicity. The present results provide evidence that hydroethanol extract of Allium sativum possess nephroprotective role and it provides protection from LPS induced nephrotoxicity. Thus is conclude that isolated compound contains nephroprotective activity against LPS induce nephrotoxicity in Swiss albino mice. [ABSTRACT FROM AUTHOR]
