Optimization of Supersaturated Solution from Stevia rebaudiana Water Extract Lead to Crystal Nucleation.

Abstract
- The nucleation of crystal growth is thought influenced by supersaturated solution. Crystallization of steviol glycoside based on the water extract of Stevia rebaudiana is challenging. We use factorial 4³D-optimal design for simultaneously optimizing supersaturation solution. As factors applied were concentrating the solution such as the addition of antisolvent, temperature and time in crystal formation. Each factor consisted of 3 levels. The supersaturated solution was observed by measurement of total dissolved solids and total suspended solids. Stevioside and rebaudioside A concentration were determined from solution which obtained in optimum condition by High Performance Liquid Chromatography (HPLC). The optimal condition of supersaturated solution was reached at a concentration of 5%, the addition of 90% ethanol as an antisolvent, a temperature of 5 °C, and a period of crystal growth for 6 hours. Optimization of supersaturation from the water extract of S. rebaudiana lead to the formation of heterogeneous primary nuclei of the crystallization process. [ABSTRACT FROM AUTHOR]
