<italic>Hymenobacter terrigena</italic> sp. nov., isolated from soil.

Abstract

A Gram-stain-negative, non-motile, non-spore-forming, rodshaped, aerobic bacterial strain, designated S1-2-2-5&lt;superscript&gt;T&lt;/superscript&gt;, was isolated from the Jeollabuk-do province, Republic of Korea, and was characterized taxonomically using a polyphasic approach. Comparative 16S rRNA gene sequence analysis showed that strain S1-2-2-5&lt;superscript&gt;T&lt;/superscript&gt; belonged to the family &lt;italic&gt;Cytophagaceae&lt;/italic&gt; in phylum &lt;italic&gt;Bacteroidetes&lt;/italic&gt;, and was most closely related to &lt;italic&gt;Hymenobacter terrae&lt;/italic&gt; DG7A&lt;superscript&gt;T&lt;/superscript&gt; (98.2%), &lt;italic&gt;Hymenobacter rubidus&lt;/italic&gt; DG7B&lt;superscript&gt;T&lt;/superscript&gt; (98.0%), &lt;italic&gt;Hymenobacter soli&lt;/italic&gt; PB17&lt;superscript&gt;T&lt;/superscript&gt; (97.7%), &lt;italic&gt;Hymenobacter daeguensis&lt;/italic&gt; 16F3Y-2&lt;superscript&gt;T&lt;/superscript&gt; (97.2%) and &lt;italic&gt;Hymenobacter saemangeumensis&lt;/italic&gt; GSR0100&lt;superscript&gt;T&lt;/superscript&gt; (97.0%). The G + C content of the genomic DNA of strain S1-2-2-5&lt;superscript&gt;T&lt;/superscript&gt; was 59.4 mol%. The detection of menaquinone MK-7 as the predominant respiratory quinone, a fatty acid profile with summed feature 3 (C&lt;subscript&gt;16:1&lt;/subscript&gt;&lt;italic&gt;ω&lt;/italic&gt;7&lt;italic&gt;c&lt;/italic&gt;/C&lt;subscript&gt;16:1&lt;/subscript&gt;&lt;italic&gt;ω&lt;/italic&gt;6&lt;italic&gt;c&lt;/italic&gt;; 32.0%), C&lt;subscript&gt;15:0&lt;/subscript&gt; iso (19.0%), and C&lt;subscript&gt;15:0&lt;/subscript&gt; anteiso (15.0%) as the major components, and a polar lipid profile with phosphatidylethanolamine as the major component supported the affiliation of strain S1-2-2-5&lt;superscript&gt;T&lt;/superscript&gt; to the genus &lt;italic&gt;Hymenobacter&lt;/italic&gt;. The DNA-DNA relatedness between strain S1-2-2-5&lt;superscript&gt;T&lt;/superscript&gt; and &lt;italic&gt;H. terrae&lt;/italic&gt; KCTC 32554&lt;superscript&gt;T&lt;/superscript&gt;, &lt;italic&gt;H. rubidus&lt;/italic&gt; KCTC 32553&lt;superscript&gt;T&lt;/superscript&gt;, &lt;italic&gt;H. soli&lt;/italic&gt; KCTC 12607&lt;superscript&gt;T&lt;/superscript&gt;, &lt;italic&gt;H. daeguensis&lt;/italic&gt; KCTC 52537&lt;superscript&gt;T&lt;/superscript&gt;, and &lt;italic&gt;H. saemangeumensis&lt;/italic&gt; KACC 16452&lt;superscript&gt;T&lt;/superscript&gt; were 49.5, 48.2, 34.1, 28.1, and 31.8% respectively, clearly showing that the isolate is not related to them at the species level. Strain S1-2-2-5&lt;superscript&gt;T&lt;/superscript&gt; could be clearly differentiated from its closest neighbors on the basis of its phenotypic, genotypic and chemotaxonomic features. Therefore, strain S1-2-2-5&lt;superscript&gt;T&lt;/superscript&gt; represents a novel species of the genus &lt;italic&gt;Hymenobacter&lt;/italic&gt;, for which the name &lt;italic&gt;Hymenobacter terrigena&lt;/italic&gt; sp. nov. is proposed.
