Abstract

Coenzyme Q10 (CoQ <subscript>10</subscript> ) is an essential part of the mitochondrial respiratory chain . Here, we describe an accurate and sensitive liquid chromatography tandem mass spectrometry (LC-MS/MS) method for determination of mitochondrial CoQ <subscript>10</subscript> in isolated mitochondria . In the assay, mitochondrial suspensions are spiked with CoQ <subscript>10</subscript> -[ <superscript>2</superscript> H <subscript>9</subscript> ] internal standard (IS), extracted with organic solvents and CoQ <subscript>10</subscript> quantified by LC-MS/MS using multiple reaction monitoring (MRM).
