Abstract

Objective To explore the expression of oncostatin M (OSM) in non-alcoholic fatty liver disease (NAFLD) and the role of OSM in promoting the development of NAFLD. Methods Thirty male SD rats were divided randomly into control group (n = 15) and model group (n = 15). Rats in control group and model group were fed with basic diet and fat-rich diet,respectively. All rats were sacrificed at the end of week 12. Fasting insulin (FINS) was measured by radioimmune methods. The ELISA double sandwich method was used to detect serum OSM concentration. The expressions of OSM and OSM receptor (OSMR) in the liver were detected by immunohistochemical method. Results Serum OSM level was increased in model group compared with control group [( 27.57士7.17)ng/mL vs. (22.64士4.52)ng/mL,P<0.05]. The expression of FINS was also higher in model group than in normal control group (3.36土0.26 vs. 3.15土0.27,P<0.05). OSM protein was expressed mainly in the cytoplasm/membrane of biliary epithelial cells and inflammatory cells,with a small amount of expression found in the cytoplasm/membrane of liver cells and Kupffer cells. OSMR was expressed mainly in the cytoplasm/membrane of the hepatocytes,especially around the central veins. The expression level of OSM and OSMR in model group was increased significantly compared with that in control group (P<0.01). Conclusion The increased expressions of OSM and OSMR in model rats may promote the development of NAFLD. [ABSTRACT FROM AUTHOR]
