Abstract

Introduction: Fully automated, and coupled with a mass spectrometric detection, on-line extraction is a very powerful and less time consuming tool than the classical extraction techniques. The objective of this study was to develop an on-line automated sample preparation using a Turboflowr® device coupled to a LC/MS/MS for measurement of methoxetamine (MXE) in human plasma. Methods: Samples (100 µL) were vortex mixed with internal standard solution (ketamine-d4 in acetonitrile). After centrifugation, 20 µL of the supernatant were injected onto a 50 × 0.5-mm C18XL column. The retained analytes were then back-flushed onto a 50 × 3-mm (3 µm) Hypersil GOLD analytical column, then eluted with formate buffer/actonitrile gradient.MXE and IS were ionized by electrospray in positive mode. Parent [M + H]+ ions were m/z 248.1 for MXE and m/z 242.1 for IS. The most intense product ion of MXE (m/z 203.1) and IS (m/z 224.1) were used for quantification. Results: The assay was accurate ([96.8% - 108.8%] range) and precise (intra- and inter-day coefficient of variation <8.8%) over the range of 2.0 (LOQ) to 1000.0 ng/mL (upper limit of quantification). No matrix effect was observed. Conclusion: Turboflowr® device coupled with LC/MS/MS allows rapid determination of compounds like MXE in plasma without extraction and without matrix effect. This method has been successfully applied to one case report after acute intoxication, showing a plasma concentration of 136 ng/mL. [ABSTRACT FROM AUTHOR]
