Abstract

The development of head and neck squamous cell carcinoma (HNSCC) is a multi-stage and multicascade phenomenon. In the signaling cascade involved in the development of HNSCC, EGF and EGFR play a key role. Several EGFR pathways have been described 1) Ras/MAP-kinase; 2) PI3K/PTEN/Akt and 3) Jak/Stat. Tumour suppressor genes are closely connected to the actors of the cell cycle. The p16/cycline D/cdk4/6 pathway is often associated with the p19/mdm2/p53 regulation pathway and these pathways are often deregulated, giving the cell significant proliferation potential. Thus biology can make a real contribution to the management of HNSCC. The potential and concrete applications are subject to knowledge of HNSCC carcinogenesis in terms of molecular epidemiology, prevention, and molecular diagnosis of relapse and developmental prognosis.
