Green Synthesis of AuNPs and Their Effect on Cell Apoptosis of HepG2 cells for Applications in Treatment of Hepatocellular Carcinoma in Nursing Care.

Abstract

Recently, metal nanoparticles synthesized by bio-fabrication methods were being used for various life-saving treatments. In our present study, AuNPs were fabricated using aqueous Panicum Maximum (P. Maximum) leaf extract. The analysis of UV–Visible spectrophotometer indicated the excitation of the formed AuNPs, specifically, a distinct SPR band positioned nearly at 530 nm. The formation of AuNPs with 45–50-nm size range was confirmed by TEM. Also, SAED pattern was utilized to characterize single crystalline nature of the biofabricated AuNPs. Further, the anticancer effects of synthesized AuNPs were studied against HepG2 cells. After incubating HepG2 cells with AuNPs, the amount of cytochrome c, CASP 9/CASP 8/CASP 3/BAX expression level, formation of ROS, and cell viability were analyzed. Outcomes of our study showed a dose determined cytotoxicity and half inhibitory concentration (IC50) of 75 μg/mL for the synthesized AuNPs. Cytotoxicity of AuNPs was accompanied by cell apoptosis and ROS induction in cell line of HepG2. Based on our results, AuNPs could have potential for development of new drugs for therapy of liver hepatocellular carcinoma in nursing care. [ABSTRACT FROM AUTHOR]
