Abstract

[Objective] The present paper aimed to investigate the expression pattern and function of two candidate lncRNAs ( lncRNA TCONS-72171 and lncRNA TCONS-62349) in bovine manunary epithelial cell with inflammation. [ Method] In the study, gene cloning and sequencing techniques were used to verify the authenticity of lncRNAs. Then, we performed sequence homology, subcellular localization, target gene prediction and KEGG functional annotation analysis. In addition, an inflanunatory response in bovine mammary epithelial cells was challenged with lipopolysaccharide ( LPS); RT-qPCR was used to detect the expression of the above two candidate lncRNAs in bovine mammary epithelial cells at 0, 3, 6 and 12 hours of LPS induction. [Result] The bovine lncRNA TCONS-72717 and lncRNA TCONS-62349 were authentic and highly conserved among some species. The expression of the above two lncRNAs were highly significantly up-regulated in LPS-induced bovine manunary epithelial cells ( P < 0. 01). LncRNA TCONS-72717 and lncRNA TCONS-62349 were mainly located in the cytoplasm and nucleus, respectively. The results of target gene prediction and KEGG functional annotation showed that the potential target genes ( LCP2, ALOX12 and ASGRJ, etc. ) of the above two lncRNAs might play a role in regulating cellular inflammation through inflammation-related signaling pathways such as JAK-STAT, NF-KB and MAPK. [Conclusion] The up-regulated lncRNA TCONS-72171 and lncRNA TCONS-62349 may play an important regulatory role in bovine manunary epithelial cells with LPS-induced inflammation, and the results may lay the foundation for an in-depth study of the molecular regulatory mechanism of dairy cow mastitis.
