基于 mt-roGFP1 荧光探针研究几种植物源除草化合物 对拟南芥根尖细胞氧化还原电位的影响.

Abstract
- [Objectives] The paper aimed to study the effects of new herbicidal compounds derived from TSC-3, Taiwanin Hand Taiwanin E on the redox potential of Arabidopsis root tip cells. [Methods] The effects of different concentrations of new herbicides on the redox potential of root cap, meristematic zone, transition zone and elongation zone of Arabidopsis thaliana were determined by using mt-roGFPI A. transgenic. [Results] Different concentrations of TSC-3, Taiwanin H and Taiwanin E could reduce the redox potential of root tip cells of the mt-roGFP1 labeled transgenic Arabidopsis thaliana plants, with the increase of the concentration of the compound, the greater the effect on the redox potential of the meristem zone of mt-roGFPI labeled transgenic A. thaliana plants. After being treated with TSC-3 at 0.1,1,5 and 10 µg mL-1 the mean values of the maximum variation in redox potential changes in root cells of the meristem zone were 13.06, 18.59, 27.74 and 29.92 mV, respectively. When treated with Taiwanin H and Taiwanin E at 1, -1 5 and 10 µg mL-1, the mean values of the maximum variation of redox potential in root cells of the meristem zone were 12.21, 15.72, 22.67 mV and 14.71, 24.03, 28.69 mV, respectively. [Conclusions] In a certain range, the greater the concentration of TSC-3, Taiwanin H and Taiwanin E, the greater of the redox potential changes in root cells of the meristem zone. It indicated that the redox potential response of A. thaliana root tip cells to these herbicidal compounds was dose-response related. Our results provide a basis for the study of the mechanism of novel plant-derived herbicides on root cell mitochondria by roGFPI fluorescence probe technology, and they also provide theoretical basis for the establishment of a rapid screening platform for herbicidal compounds.
