高尿酸血症大鼠睾丸组织中差异表达的 lncRNA 及意义初步探讨.

Abstract

Objective To identify differentially expressed long non-coding RNAs(lncRNA) in testis of hyperuricemia-induced male infertility rats. Methods Hematoxylin-eosin(HE) staining was used to observe the histopathological morphology of rat testis. RNA was extracted from the testis of male SD rats in control group and hyperuricemia group and sequenced. Differentially expressed lncRNA were analyzed between two groups, and their target genes were predicted. GO and KEGG functional enrichment analysis were performed for differential lncRNA and target mRNA. Results Compared with those in the control group, 89 differentially expressed lncRNA(42 up-regulated and 47 down-regulated) and 50 differentially expressed mRNAs(29 up-regulated and 21 down-regulated) were identified in hyperuricemia group. GO enrichment analysis revealed that these differentially expressed lncRNA were mainly associated with the regulation of insulin receptor signaling pathway, the regulation of inflammatory response, developmental process and G-protein-coupled glutamate receptor binding. KEGG pathway enrichment analysis showed that differentially expressed lncRNA were mainly enriched in insulin signaling pathway, cGMP-PKG signaling pathway, cAMP signaling pathway and calcium signaling pathway. Conclusions The differential expression of lncRNA in testicular tissue of HUA rats may help elucidate the potential mechanism of lncRNA in hyperuricemia-induced reproductive disorders in male rats.
