小檗碱对幽门螺旋杆菌感染小鼠胃粘膜损伤和免疫功能的影响机制研究.

Abstract

Objective: To investigate the effect of berberine on gastric mucosal injury and immune function in mice infected with Helicobacter pylori. Methods: Fifty SPF male mice were randomly divided into 5 groups, including blank control group, Helicobacter pylori model group, berberine low-dose group (25 mg/mL, group A), medium-dose group (50 mg/mL, group B) and high-dose group (100 mg/kg/d, group C). Rats in blank group were intraperitoneally injected with normal saline of equal volume, and the other groups were orally administered Helicobacter pylori to establish a mouse model of Helicobacter pylori infection. The survival rate and apoptosis rate of gastric mucosal epithelial cells were detected, and the levels of apoptotic protein, inflammatory factors, immune function and IL-4/STAT6 mRNA of gastric mucosal epithelial cells were measured. Results: The survival rate of gastric mucosal epithelial cells in model group, berberine low-dose, medium-dose and high-dose groups was significantly lower than that in blank control group (P<0.05). With the increase of berberine dose, cell survival rate was gradually increased (P<0.05). The apoptosis rate of gastric mucosal epithelial cells in model group, berberine low-dose, medium-dose and high-dose groups was significantly higher than that in blank control group (P<0.05). With the increase of berberine dose, the apoptosis rate decreased gradually (P<0.05). Compared with model group, the levels of apoptotic protein Bax and Cl-caspase-3, TNF-α, IFN-γ, IL-10 and TGF-β of mouse gastric mucosal epithelial cells in berberine low, medium and high dose groups were significantly decreased (P<0.05), and with the increase of berberine dose, The levels of Bax and Cl-caspase-3, TNF-α, IFN-γ, IL-10 and TGF-β were decreased (P<0.05). Compared with model group, the levels of CD3+, CD4+ and CD8+, IL-4 mRNA and STAT6 mRNA in the supernatant of berberine low-dose, medium-dose and high-dose groups were significantly increased(P<0.05), and with the increase of berberine dose, CD3+, CD4+ and CD8+ levels, IL-4 mRNA and STAT6 mRNA were increased gradually(P<0.05). Conclusion: Berberine can alleviate gastric mucosal damage and enhance immune function in mice infected with Helicobacter pylori, which may be related to IL-4/STAT6 signaling pathway.
